| In this study,the quality of medicinal Mulberry from Aksu,Kashgar and Turpan in different regions of Xinjiang was compared and analyzed,final selection taking Xinjiang Kashgar medicinal mulberry and Cabernet Sauvignon grape as raw materials,a nutritious and healthy medicinal mulberry grape wine was prepared by optimizing the fermentation conditions to improve the preservation rate of bioactive substances in medicinal mulberry and grape.The in vitro test of the composite wine was designed,combined with animal test,to study and analyze the in vitro antioxidant activity and in vitro binding cholate,And observe its effects on lipid metabolism,oxidative stress,organ and tissue lesions in hyperlipidemia model rats.The specific research results are as follows:(1)The water content,total acid,soluble solids,p H value,crude protein,total phenol,total flavonoids,vitamin C,anthocyanin content,total free amino acid content and mineral content of medicinal mulberry in Aksu,Kashgar and Turpan were analyzed for principal component analysis.The result is as follows:Moisture content between75.5%-80.21%,p H value between 2.8-3.5,soluble solids content between16.5%-20%,total acid content between 20-70 mg/g,crude protein content between 22.26-47.45 mg/g,total phenolic content is between 35.40-41.40 mg/g,the total flavonoid content is between 16.35-21.52 mg/g,the anthocyanin content is between 302.4-358.5 mg/100g,and the vitamin C content is between 37.85-45.6mg/100g,The average value of mineral content from high to low is:Turpan medicinal mulberry>Kashi medicinal mulberry>Aksu medicinal mulberry,The average value of total free amino acid content from high to low is:Kashi medicinal mulberry>Turpan medicinal mulberry>Aksu medicinal mulberry.The comprehensive scoring results of medicinal mulberry from the three origins are:Kashi medicinal mulberry>Turpan medicinal mulberry>Aksu medicinal mulberry,indicating that from the perspective of principal component analysis of various physical and chemical indicators,minerals,bioactive substances and amino acids,the quality of Kashi medicinal mulberry is better protrude.(2)Taking Xinjiang medicinal and food homologous plant medicinal mulberry and Xinjiang wine grape Cabernet Sauvignon as raw materials,a single factor experiment was designed combined with response surface analysis to optimize the optimal fermentation conditions when the content of total phenols and total flavonoids in the medicinal mulberry wine was maximum.The optimum process conditions were finally obtained as follows:the inoculum amount is 0.3%,the fermentation time is 8 d,the fermentation temperature is 26℃,and the mass ratio of the raw materials of grape and medicine mulberry is 1:3.The total phenolic and total flavonoid contents of the obtained medicine mulberry grape wine were 2872.48mg/L and 1763.29 mg/L.Fermentation under these conditions yielded high content of total phenols and total flavonoids,harmonious aroma of mulberry and grape,balanced taste,strong sense of wine body structure,and outstanding typicality of mulberry wine.(3)Three antioxidant systems(DPPH,ABTS~+,·OH)were used to evaluate the antioxidant activity of medicine mulberry grape wine.The in vitro hypolipidemic effect of medicine mulberry grape wine was evaluated by the binding ability of different concentrations of medicine mulberry grape wine with glycocholate and taurocholate.The results showed that the scavenging rates of DPPH,ABTS~+and hydroxyl radicals were 98.83%,98.38%and 99.24%,when the Volume concentration of the Medicine Mulberry Grape Wine was the highest.respectively;the combination with glycocholate and taurocholate The rate reached 69.36%and 73.28%respectively.The results indicated that the medicine mulberry grape wine had stronger antioxidant activity and better in vitro hypolipidemic activity compared with similar fruit wines.(4)The hyperlipidemia model of rats was established by feeding high-fat diet,and at the same time,intragastric administration was performed.After 8 weeks,the contents of TC,TG,HDL-C,LDL-C,SOD,CAT,and MDA in rat serum were determined by kits.The pathological sections of Wistar rat liver and heart were stained with HE and the degree of liver and hear lesions of hyperlipidemia rats was observed under microscope.In animal experiments,medicine mulberry grape wine significantly reduced the contents of TC,TG,LDL-C and MDA in Wistar rats,and significantly increased the contents of high-density lipoprotein HDL-C,SOD and CAT,and improved serum Lipid peroxidation,which reduces the risk of arteriosclerosis.Liver pathological examination found that compared with the hyperlipidemia model group,the steatosis of liver pathological sections of Wistar rats treated with medicine mulberry grape wine was significantly improved.It shows that the medicine mulberry grape wine can effectively improve the liver tissue and inhibit the accumulation of lipids in the liver. |
