Polyphenols are a class of phenolic hydroxy compounds that are widely present in plants.They have the functions of anti-oxidation,scavenging free radicals,inhibiting cancer,lowering blood sugar and blood lipids,delaying aging,and enhancing immunity.They have high application and development value.In this study,We take Xinjiang ‘Jinghe Lycium barbarum L’ as the research material.,The extraction process parameters of polyphenols from Lycium barbarum was studied by ultrasonic-assisted extraction method,and the antioxidant activity of Lycium barbarum was evaluated,An ultra performance liquid chromatography-ion mobility quadrupole-time-of-flight mass spectrometry(UPLC-IM-Q TOF MS)method was established Identification and identification of polyphenols in Lycium barbarum extract and the main polyphenols were quantitatively analyzed,which provided important reference for the study of active phenols in lycium barbarum.The study draws the following main conclusions:1.The factors affecting the extraction of Lycium barbarum polyphenols were as follows: solid-liquid ratio> extraction temperature>methanol extraction liquid fraction>extraction time.After response surface optimization,the extraction conditions of Lycium barbarum polyphenols were as follows: solid-liquid ratio1:21 g/m L,methanol extraction liquid fraction 60%,extraction temperature 50 ℃,extraction time 39 min.2.The content of total phenols and total flavonoids in Lycium barbarum in Ningxia and Jinghe production areas are significantly different(P<005);when the same variety of Lycium barbarum(Ningqi 7#)is planted in different production areas,and the content of total phenolics and total flavonoids in Jinghe Lycium barbarum is higher than that in Ningxia.In the same production area(Jinghe production area),the comparative analysis of different varieties of Lycium barbarum,Ningqi 7# Lycium barbarum has the highest content of total phenols and total flavonoids;for polyphenols in Lycium barbarum,all varieties of Lycium barbarum are the highest in autumn fruit Lycium barbarum.In summer fruit Lycium barbarum.3.There are differences in the antioxidant activities of Lycium barbarum in Ningxia and Jinghe production areas;the antioxidant activities of Lycium barbarum in the same production area(Jinghe production area)and different varieties(summer/autumn fruit)are different.Lycium barbarum Ningqi 7#(autumn fruit)has thestrongestabilitytoscavenge2,2-diphenyl-1-picrylhydrazyl(DPPH)freeradicalsandtotalreducingpower,and Lycium barbarum1801#(autumnfruit)hasthestrongestabilitytoscavenge2,2’-azino-bis-3-ethylbenzothiazoline-6-sulfonicacid(ABTS)freeradicals,indicating that there are significant differences(P<005)in the antioxidant activity of different varieties of Lycium barbarum in the same region;The three evaluation methods of DPPH radical,ABTS radical scavenging ability and total reducing force were combined with the comprehensive analysis of the correlation with the polyphenol content of Lycium barbarum,the antioxidant activity of Lycium barbarum was significantly positively correlated with the content of polyphenols(P<0.05),indicating that the content of polyphenols in Lycium barbarum had an important influence on the antioxidant activity.4.In this study,to analyze the phenolic composition of Lycium barbarum,Using UPLC-IM-Q TOF MS technology to identify and analyze Lycium barbarum polyphenols,a total of 28 polyphenols were identified,including 18 flavonoids,8 phenolic acids,2 tannic acids,indicating that Lycium barbarum contains a large number of polyphenols,and mainly composed of flavonoids.5.The main polyphenol components of Lycium barbarum are: rutin,morin,cinnamic acid,isoquercetin,p-coumaric acid,ferulic acid,kaempferol-3-rutinoside,isorhamnetin,Isorhamnetin-3-O-rutinoside;the contents are in the following order: rutin>p-coumaric acid>kaempferol-3-rutinoside>isoquercetin>isorhamnetin>Isorhamnetin-3-O-rutinoside;Jinghe Lycium barbarum rutin and p-coumaric acid content were significantly higher than Ningxia Lycium barbarum,indicating that rutin and p-coumaric acid are the most important polyphenols in Jinghe Lycium barbarum currently identified.polyphenolic compounds.
