Hollow mesoporous silica had hollow cavity and mesoporous layer,which had great potential in the field of drug carrier.Different responsive drug carriers were prepared by coating the outer layer of functional composites on hollow mesoporous silicon.Hollow mesoporous silicon was prepared by hard template method,reacted with silane coupling agents MPTMS and APS,and introduced amino and sulfhydryl functional groups.Finally,the reduction response carrier HMSN-ss-CD was prepared by sulfhydryl disulfide bond exchange reaction;The carboxyl group was modified to the surface of hollow mesoporous silicon by the reaction of amino and succinic anhydride.The copolymer of NHMA and NIPAM was coated on the surface of hollow mesoporous silicon by free radical polymerization,and the carrier HMSN(-COOH)@P(NIPAM-co-NHMA)with p H and temperature response was synthesized.HMSN-ss-CD was adsorbed at 30℃for 24 hours,and HMSN(-COOH)@P(NIPAM-co-NHMA)was adsorbed at 25℃for 12 hours.The adsorption effect of the carrier was the best.The saturated adsorption capacity was 62.3 mg·g-1and70.58 mg·g-1,the drug loading rate was 5.76%and 8.73%,and the entrapment efficiency was 43.67%and 53.14%.The adsorption process of the carrier conforms to Langmuir isothermal adsorption and quasi second-order kinetic model.In vitro drug release studies showed that the drug release rate of QUE@HMSN-ss-CD was only 8.1%in phosphate buffer solution without glutathione,while the drug release rate could reach 89.05%when the carrier was in PBS solution containing 20 m M GSH,indicating that the drug sustained-release system had good redox response.The drug release rate of QUE@HMSN(-COOH)@P(NIPAM-co-NHMA)in the environment with p H value of5.5 and temperature of 40℃was 12.33%,indicating that the drug sustained-release system had good temperature and p H sensitivity.The release of QUE by the drug loading system conforms to the Korsmeyer-Peppas model.Hemolysis experiment confirmed that the two carriers had good biosafety.Figure 39;Table 10;Reference 70...
