Objective: A polysaccharide buccal tablets of Polygonatum cyrtonema Hua was developed,which has a convenient and rapid preparation process and can regulate the blood sugar and intestinal flora of type Ⅱ diabetes mice.Methods: Study on the extraction method of polysaccharides from polygonatum cyrtonema Hua with ultrasound assisted extraction,and the best extraction conditions were determined on the basis of single factor and response surface methodology.Using the extracted Polygonatum cyrtonema Hua Polysaccharides as raw materials and adding auxiliary materials to develop a polysaccharide buccal tablets.According to the comprehensive score of the taste,appearance and compression of the buccal tablets,the response surface method was used to screen the formula of the buccal tablets and determine the best ratio.Finally,the effects of Polygonatum cyrtonema Hua polysaccharide buccal tablets on regulating blood sugar and intestinal flora in mice with type Ⅱ diabetes were verified.At the same time,the diversity and abundance of intestinal flora in type Ⅱ diabetes mice were deeply analyzed by high-throughput sequencing technology.Results:(1)The optimum technological conditions for polysaccharide extraction were as follows: ultrasonic time 49 minutes,ultrasonic temperature 71 ℃,solid-liquid ratio 1:40,ultrasonic power 270 W,After verification,the average mass fraction of polysaccharides is83% and the yield was 25.07% ± 0.15%.(2)The best formula of the Polygonatum cyrtonema Hua buccal tablets is as follows: Polysaccharide powder addition 60%,the addition amount of filler is 19.91%(mannitol: microcrystalline cellulose=3:1,mass ratio),the addition amount of flavor correction agent xylitol is 14.55%,the addition amount of flavor correction agent citric acid is 5.04%,the addition amount of lubricant magnesium stearate is 0.5%,and the volume fraction of granulating wetting agent ethanol is 85%.(3)Investigation on the forming quality of buccal tablets:The surface of Polygonatum cyrtonema Hua polysaccharides buccal tablets is smooth,without color spots or cracks.The preparation process has good viscosity,fluidity,and compressibility,which can meet the needs of industrial production.The weight of Polygonatum cyrtonema Hua polysaccharides buccal tablets is about 0.6g/tablet,and the polysaccharide content of Polygonatum cyrtonema Hua polysaccharides buccal tablets is not less than 0.28g/tablet.According to the recommended intake in the Pharmacopoeia for the human body,it is recommended to take three tablets a day,three at a time.(4)Study on regulating intestinal flora: The effects of high dose(PBT_H),medium dose(PBT_M)and low dose(PBT_L)of buccal tablets on the flora of mice were investigated by using the model of type Ⅱ diabetes mice,and the model control group(T2DM),acarbose positive control group(Acar)and blank control group(NFD)were set up for comparative study.The results showed that compared with T2DM group,oral buccal tablet could significantly reduce the blood glucose level of mice after 5 weeks(P<0.05).In the glucose tolerance test,the blood glucose level of T2DM group increased rapidly after intragastric administration of glucose solution and remained at high blood glucose level within 2 hours,while the blood glucose level of NFD group remained basically unchanged.And the Acar group,PBT_L group,PBT_M group,PBT_H group compared with the T2DM group,the blood glucose level decreased significantly(P<0.05)after 1 hour of gavage,and it was dose-dependent.The experiment found that polysaccharide buccal tablets can change the composition and structure of the microbiota.Firmicutes and Bacteroidota are the two main bacterial phyla that can be regulated by polysaccharide buccal tablets in the intestine.Polysaccharide buccal tablets significantly change the relative abundance of Firmicutes and Bacteroidota.The independent relative abundance and the relative abundance of Firmicutes/Bacteroidota ratio of PBT_H group recovered after intervention compared with T2DM group(P<0.05).This study proved that the test dosage group of Polygonatum cyrtonema polysaccharide buccal tablets had a significant reduction effect on Lachnospiracea and Desulfovibrionacea(P<0.05).The results also showed that PBT_H group could significantly increase the content of Prevotellacea(P<0.05).Conclusion: A convenient and fast preparation process of Polygonatum cyrtonema Hua polysaccharide buccal tablets was successfully developed.The buccal tablets can regulate the increase of blood sugar and intestinal flora in type Ⅱ diabetes mice.
