Berberis heteropoda schrank,berberidaceae berberis L perennial thorny deciduous shrub,mainly distributed in the west and southwest of China.As a wild plant of medicinal and food homology,it mainly has the effect of clearing heat,anti-inflammatory,quenching thirst and stopping diarrhea.The shortage of medicinal sources and ecological damage caused by insufficient medicinal plant resources and growth retardation have limited the use of blackberry barberry plants.The discovery that endophytic fungi in medicinal plants can synthesize medicinal active substances that are identical or similar to host plants solves the problem of insufficient medicinal plant resources and ecological destruction.Endogenous fungi of medicinal plants have the advantages of easy culture,easy control,fast growth and low requirements for environmental space,and their potential value is mainly reflected in the research and development and application of new drugs.Plant endophytic fungi are diverse,can be metabolized to produce different kinds of compounds,may metabolize the production of unsupervised compounds,and are one of the sources of active natural metabolites with novel structures.The true bacteria in medicinal plants have anti-inflammatory,anti-tumor and other activities,and are a treasure trove of natural active substance resources.According to research reports,at present,the research on blackberry barberry is mainly in the plant aspect,and there are fewer studies on the endophytic fungal species,bioactive ingredients,and corresponding drug efficacy relationships of blackberry barberry.In this study,through previous literature screening,it was found that Berberis has good biological activities such as anti-tumor,anti-virus,anti-inflammatory and anti-oxidation.In addition to being used as folk medicine and jam,a large number of resources have not been developed and utilized.In order to improve this situation,mining the economic value of berberis barberis,this paper selects the medicinal plant Berberis barberis as raw material,from which the isolation and purification of endophytic fungi,the chemical composition and anti-inflammatory activity of secondary metabolites were studied,the main research content is as follows:Endophytic fungi were screened from the blackberry barberry stem and leaf plants,and the isolated monomeric compounds were actively detected.[Methods]Tissue block culture method and plate scribing method were used to isolate and purify endophytic fungi of blackberry barberry plants,endophytic fungi were preserved by slope cryopreservation method,endophytic fungi were identified by morphological methods and molecular biology,and ethyl acetate parts of the fermentation extract of N1 strain of Fine Branch Sporangia were systematically isolated by MCI,silica gel,Sephadex LH-20 and other modern chromatography techniques To evaluate the anti-inflammatory activity of monomer compounds by establishing a lipopolysaccharide(LPS)-activated RAW264.7 macrophage inflammatory model.The structures of the compounds identified by ultraviolet,infrared,mass spectrometry,nuclear magnetism and other technologies and compared with the literature data are:2-(2-hydroxypropyl)-6-methyl-2,3-dihydro-4H-pyran-4-one(1),(2Z,4E,6E)-7-(3,4-dihydroxy-2,6,8-trimethyldecahydronaphthalen-1-yl)-2-methylhept a-2,4,6-trienoic acid(2),(2S,4R,4a S,5S,6R,8a R)-6-ethylhexahydro-2H-2,5-epoxychromene-4,4a(5H)-diol(3),(8R,8a S)-8-ethyl-5-hydroxy-6,7,8,8a-tetrahydro-2H-chromen-2-one(4),(4S,4a R,5S,6R)-6-ethyl-3,4,6,7-tetrahydro-2H-chromene-4,4a,5(5H)-triol(5),Orcinotriol(6),4-Hydroxyphenethyl alcohol(7),α-acetylorcinol(8),Adidadienoic acid(9),4-Hydroxybenzaldehyde(10),2,4-Dichlorobenzoicacid(11).Of these,Five of the compounds were unreported and six were known.In summary,this paper obtains 9 endophytic fungi from the blackberry barberry plant by isolation and purification,ferments 1 of the endophytic fungi in large quantities,and identifies 11 monomeric compounds,5 of which are new,and activity screening for compounds 1 to 11.This paper has a certain reference value for the comprehensive utilization of plant resources and anti-inflammatory drug research and development of endophytic fungi from Berberis heteropoda schrank.
