| Chronic wounds are known as "silent epidemics",which afflict tens of thousands of patients.Due to the complexity of the microenvironment of chronic wounds,wound healing is often not ideal.Therefore,how to effectively treat chronic wounds and improve the recovery effect of chronic wounds is currently an urgent problem to be solved.Platelet derived growth factor(PDGF)plays an important role in cell proliferation,migration,and differentiation,and can effectively promote chronic wound healing.However,PDGF is susceptible to the proteolytic environment in chronic wounds,leading to issues such as short half-life and easy inactivation,making it unable to maintain stable concentrations at the wound site for a long time.Frequent repeated injections or prolonged repeated administration at physiological doses to maintain its concentration at the wound site can lead to serious side effects.Therefore,developing precise and controllable PDGF delivery strategies with long-term effects is crucial for chronic wound repair.In this paper,hyaluronic acid(HA)was modified to prepare methacrylic acid(MA-HA),and a UV induced p H responsive degradable hydrogel(MAHA-Gel/PDGF)loaded with PDGF was constructed for the treatment of chronic wounds.The release rate of PDGF encapsulated in the hydrogel can be precisely controlled by adjusting the proportion of crosslinkers with different p H values responding to bond breaking in the hydrogel.In vitro cell experiments showed that MA-HA-Gel/PDGF hydrogel had good biocompatibility,and PDGF released from the hydrogel could promote fibroblast migration.The experimental results in vivo further proved that in the full-thickness wound model of diabetes mice,MA-HA-Gel/PDGF hydrogel can promote granulation tissue maturation,collagen deposition and angiogenesis,effectively promoting the process of chronic wound healing.In the future,we hope to further research and explore this hydrogel system,so that it can also be used as a delivery carrier for other growth factors or drugs,so as to play a related therapeutic role,broaden the application field,and make this delivery system better serve other types of diseases. |
