Natural Resveratrol(Res)has a variety of physiological activities,such as antioxidant,anti-tumor,and prevention of diabetes.It has great research value in the fields of food and pharmacy.At present,there is a lack of research on the refining,separation,and application system of peony seed meal.For the reuse of peony seed meal resources,this study used local peony seed meal as raw material to conduct research on the extraction of Res from peony seed meal,and used ultrasound to extract Res from peony seed meal.At the same time,the separation,purification and identification of Res from tree peony seed meal were carried out,and finally it was prepared into solid dispersion(SD),in order to improve the bioavailability of Res from tree peony seed meal,and provide theoretical basis and technical support for the comprehensive development and utilization of oil tree peony seed resources.The research results were as follows:Using ultrasonic assisted extraction method,the extraction rate of Res from peony seed meal was used as an indicator to study the effects of ethanol concentration,ultrasonic power,and ultrasonic treatment temperature on the extraction rate of Res.After response surface optimization,the optimal extraction process conditions were obtained as follows:extraction temperature 60℃,ethanol concentration 70%,and ultrasonic power 120 W.At this point,the actual yield is 2.83%,providing experimental basis for the next step of research.At the same time,this thesis optimized the purification process of macroporous resin on peony seed meal extract,enriches Res with macroporous resin,and compares the treatment effects of eight macroporous resins through adsorption and analysis experiments on peony seed meal Res in view of the physical and chemical properties of Res,and selects DA-201 macroporous resin for the next experiment.The optimal static analysis conditions were determined by the static adsorption and analytical process parameters:p H5 on the crude extract of Danseed meal,the resolution time was 190 min,and the ethanol concentration of the eluent was 70%.The dynamic adsorption-analysis conditions were determined by the dynamic adsorption and analysis process parameters:the concentration of the upper column solution was 0.8 mg/m L,the p H of the crude extract of peony seed meal was 5,the loading flow rate was 0.4 m L/min,and the loading amount was 84 m L.The ethanol concentration of the eluent was 70%,and the resolution flow rate was 0.8 m L/min.Under these conditions,the enrichment of Res in peony seed meal was studied.To further further improve the purity of peony seed meal Res,a purity of 98.32%of peony seed meal Res could be obtained through Prep-HPLC purification,which was helpful for later research on its formulation.The thin layer chromatography results only showed a purple red spot on the corresponding position of the reference substance chromatography in the test substance chromatography.The thin layer chromatography chromatograms of Res obtained from purified peony seed meal and Res standard showed that R_f was basically the same.Finally,purified Res was used as raw material to prepare SD,and carboxymethyl chitosan(CMCS)and polyvinylpyrrolidone(PVPK29/32)were selected as joint carriers to prepare ternary SD.The optimal formulation of ternary SD was obtained by optimizing the cumulative dissolution.The FITR results indicate that CMCS can form intermolecular hydrogen bonding interactions with PVP and drugs,respectively.The XRD and MDSC results indicate that the addition of CMCS can improve the compatibility of various components in the mixed system.CMCS can improve the solubility of drugs in the carrier,and CMCS can increase the Tg of the mixed system,effectively reducing the molecular migration rate.The stability test results of the optimal formulation showed that the Res ternary SD prepared by this process had good stability,indicating that PVPK29/32 and CMCS had good application prospects as carrier materials. |
