China is abundant in natural peat resources,ranking sixth in the world.Woody peat as one of the important types of peat,its carbon content is high,generally up to 60%-65%,and its humic acid and organic matter content is also relatively high with porous structure.It is a natural organic fertilizer with feature of preserving moisture and fertility can be applied to plant growth conditioner,soil conditioner and organic inorganic compound fertilizer manufacturing,etc.Trichoderma spp.as an important plant rhizosphere growth-promoting fungus is widely found in the environment and has many beneficial functions for plant health.It can widely used in agricultural biological control,food industry,feed production,bio-genetic engineering technique and so on.Solid fermentation is easy to operate with feature of low cost,low carbon and pro-environment.The culture medium is relatively simple,the fermentation conditions are simple and efficient,but it can obtain high yield and quality fungal spores,and the production process has little impact on the environment.In this study,woody peat was used as the main raw material for solid fermentation of Trichoderma spp.Firstly,the parameters of solid fermentation were determined by proportioning experiment of woody peat and different materials(Rice straw,rice bran and stevia residue),then the solid fermentation process was optimized to obtain biological fertilizer products with both high amount Trichoderma spp and high level of humic acid concentration.The main research results are as follows:1.Subsidiary material were screened when the main material is woody peat for solid-state fermentation of Trichoderma.Results showed that the effect of fermentation of Trichoderma is the best with equal proportion of woody peat and rice bran.The specific parameters is as follow:adding 3%amino acid solution,115℃sterilization 60 min,access 10%Trichoderma viride liquid fermentation medium,fermentation material about 2cm thick,on average,28℃,7 days fermentation under the condition of illumination,efficacious living-cells can reach 6.67×108cfu/g.2.The orthogonal optimization experiment further optimized the core fermentation parameters.When the ratio of rice bran and woody peat was 2,the inoculation amount was7.5%,and the fermentation period was 7 days,efficacious living-cells of Trichoderma solid fermentation was the highest,which reached 8.13×108cfu/g.After 7 days fermentation,the total humic acid content of fermentation products was increased from8.5%to 16.57%,with an increase of 94.94%.The structural changes of substance in raw material with different fermentation periods were measured by infrared spectroscopy.Results showed that the absorption peaks of starting crude before and after fermentation were similar,and the characteristic peaks were mainly at 427,471,533,695,801,911,1003,1028,1097,1271,1402,1508,1611,2851,2919,3026,3126,3623,3651,3706 cm-1.We futher made a comparative analysis based on these data,found that carbohydrate,lignin and aliphatic compounds-CH3,-CH2and-CH decrease after 5 days fermentation,indicated that cellulose,hemicellulose,and lignin are decomposed into gradually,and accompanied by the generation of aromatic compounds.At the late fermentation stage(7 days),due to the rapid reproduction of Trichoderma spp,a large number of metabolites were secreted,leading to the enhancement of the relative intensity of the corresponding absorption peaks of carbohydrates,lignin and fat compounds.3.Chinese cabbage green house experiments confirmed that chlorophyll,leaf number,aboveground/underground part fresh weight,the dry weight and leaf area increased in the treatment of Trichoderma biological organic fertilizer with chemical fertilizer in half,indicated the potential of Trichoderma biological fertilizer probably improve fertilizer utilization efficiency,result in the promotion of the growth of Chinese cabbage.
