Astragalus membranaceus is a genuine medicinal material of Inner Mongolia which is also a kind of homologous material of medicine and food,containing polysaccharides,flavonoids,alkaloids and other active substances,which can regulate immunity,antioxidant,anti-tumor,antibacterial,antiviral,hypoglycemic and other effects.In this study,astragalus polysaccharide(APS)was extracted by compound enzymes,purified by D101 macroporous resin,and the composition,antioxidant and anti-tumor activities of the polysaccharide were studied.The astragalus polysaccharide complex drink was developed.The main research results are as follows:(1)Extraction and purification of astragalus polysaccharide.The bset extraction means and technological conditions of astragalus polysaccharides were confirmed by single factor and orthogonal experiment.Compound enzyme method was used,the ratio of astragalus powder to liquid was 1﹕15,the extraction was carried out at 65℃ for 120 min.The yield of polysaccharides was 35.65±0.216% and the polysaccharide content was28.73% by 6 compound enzymes extraction,which were cellulase,hemicellulase,xylanase,pectinase,glucose oxidase and papain.The astragalus polysaccharide was purified by macroporous resin static adsorption method.The D101 macroporous resin was 1.2 g/m L,the adsorption time was 80 min,the rotation speed was 110 r/min,and the content of astragalus purified polysaccharide was 61.1%.The purified astragalus polysaccharide was verified as acidic polysaccharide by infrared spectrum scanning,which contained pyranose.(2)Antioxidant activity of astragalus polysaccharides in vitro.The DPPH free radical scavenging ability of APS was 24.45% when the concentration of APS was 0.6mg/m L,which was 0.48 times that of Vc.The iron ion reducing capacity was 22.4%when the concentration of APS was 0.8 mg/m L,which was equal to 0.5 times of Vc.When the concentration of APS was 0.6 mg/m L,the hydroxyl radical scavenging ability of it was 71.76%,which was 0.67 times that of Vc.The FRAP value of APS was 8.52%at 0.6 mg/m L,which was 0.5 times that of Vc.(3)Effects of astragalus polysaccharide on pancreatic cancer cells and spermatogonium.The pancreatic cancer cell experiment showed that APS inhibited the proliferation of pancreatic cancer cells,and astragalus pure polysaccharide had better effect than astragalus crude polysaccharide.When the concentration of astragalus pure polysaccharide was 0.8 mg/m L,the mortality rate of pancreatic cancer cells reached56.67±5.774%.The spermatogonial cell experiment showed that APS could promote the proliferation of spermatogonial cells,and the effect of pure astragalus polysaccharide was better than that of crude astragalus polysaccharide.When the concentration of pure astragalus polysaccharide was 1.6 mg/m L,the death rate of spermatogonial cells was the minimum of 1.67±1.155%.(4)Development and formulation optimization of astragalus polysaccharide compound drink.The optimal formula of APS compound drink was determined by single factor and response surface experiment: Astragalus polysaccharide content was 60%,black tea content was 30%,honeysuckle content was 0.25%,stevioside and erythritol was3%,D-isoascorbate sodium was 0.1%.There were 77.9 mg tea polyphenols,15.6 mg caffeine,and 0.1 g carbohydrate in 100 m L APS compound drink.Astragalus polysaccharide compound drink is a fresh,pure,moderate taste,sugar free healthy drink.
