| Tremella has five main types of polysaccharides: acidic heteropolysaccharides,acidic oligosaccharides,neutral heteropolysaccharides,cell wall polysaccharides,and extracellular polysaccharides.While most tremella research focuses on dry tremella,few studies have been conducted on ice fresh tremella.Therefore,this study used ice fresh tremella to extract tremella polysaccharides,and then purified and applied them in eye cream according to the concepts of green chemistry and modern cosmetics.The research includes the following contents:Optimization of extraction process of polysaccharides from tremella: The hot water extraction method was used to extract polysaccharides from ice fresh tremella.The effects of different solid-liquid ratios,extraction temperatures,and extraction times on polysaccharide content were investigated,with polysaccharide content as the index.A response surface test of 3 factors and 3 levels was established by setting different factor levels.Through the analysis of the regression model,the optimal process conditions were obtained: the ratio of material to liquid was 1:20,the extraction temperature was95°C,and the extraction time was 1.6 h.The optimum process was verified by repeated experiments three times,and the polysaccharide contents were 6.74%,6.72%,and6.75%,respectively,all between(6.73 ± 0.02)percent,which was consistent with the predicted value of the model,indicating that the model results were stable.It is feasible to extract tremella polysaccharide using this process condition.Purification of tremella polysaccharides: Taking a green chemistry approach,polyamide column chromatography was utilized to remove protein.The crude tremella polysaccharide solution was obtained through hot water extraction.The solution was concentrated to a certain level and anhydrous ethanol was added to precipitate the polysaccharides while keeping the ethanol concentration between 70% and 80%.The content of crude polysaccharide after water extraction and alcohol precipitation was found to be 88.76%,with an average yield of 4.84%.To investigate the effects of sample volume,sample volume,adsorption equilibrium time,and elution rate on the total sugar retention rate and deproteinization rate,these parameters were evaluated using these indicators.The optimum elution process was determined based on the total sugar retention rate and deproteinization rate,with a sample volume of 60 mg,sample volume of 30 m L,adsorption equilibrium time of 30 min,and elution rate of 3 m L/min.The purified tremella polysaccharide had a total sugar content of 97.90%,with a yield of high-purity tremella polysaccharide of 75.72%.The infrared spectrum scanning of the purified material showed that the characteristic absorption peak was consistent with that of pure tremella polysaccharide,indicating that the prepared sample was a high-purity tremella polysaccharide.Preparation and efficacy of tremella polysaccharide eye cream: The in vitro hygroscopicity and moisture retention of the prepared tremella polysaccharide were investigated,showing that the tremella polysaccharide had an efficient moisturizing effect.The purified high-purity tremella polysaccharide was applied to eye cream,and natural animal and vegetable oils were used as the basic raw materials of oiliness.The emulsifier was chosen based on the HLB value,and the eye cream matrix was tested for its p H value,heat resistance,cold resistance,heat and cold resistance alternating test,and centrifugal test.The best emulsifier formula was found to be 2.5% Tween-80 and1.5% A-165 emulsifier.Carbopol-940 and xanthan gum were selected as the thickening system for the eye cream,with a sensory evaluation test(9-point method)used to determine the best ratio of 0.1% Carbopol-940 and 0.2% xanthan gum.An anti-corrosion system of eye cream was established using a combination of phenoxyethanol and 1,2-pentanediol,which was tested for efficiency using the CTFA method in a microbial challenge test.The best anti-corrosion compound system was found to be 0.4% phenoxyethanol and 2% 1,2-pentanediol.To determine the optimal concentration of tremella polysaccharide,the change rate of human skin water content was measured using different concentrations of polysaccharide as a variable.The optimum addition amount of tremella polysaccharide was found to be 0.15%.Using this formula,tremella polysaccharide eye cream was prepared and was found to be refreshing,non-greasy,smooth,and easy to spread,with an efficient moisturizing effect.Overall,this paper focuses on green chemistry by using iced fresh tremella as a raw material,optimizing the process of hot water extraction of polysaccharides,and using the polyamide chromatography column method to purify tremella polysaccharides to obtain high-purity polysaccharides.The study concludes that tremella polysaccharide eye cream is an excellent formulation with a good texture and an efficient moisturizing effect. |
