| How to realize the resolution of chiral drugs and the screening of antioxidant peptide chains by chromatography is a research hotspot in recent years.Porous poly(styrene-divinylbenzene)microspheres are widely used as column fillers because of their high osmotic pressure and high specific surface area.At present,the most common compounds with chiral recognition sites in chiral resolution chromatographic columns include polysaccharides,crown ethers and cyclodextrins.However,the chiral recognition ability of these compounds to a variety of chiral drugs is insufficient,so functional modification is introduced.In order to expand the application of chromatography and improve the resolution of peptide library by reversed-phase high performance liquid chromatography,it is necessary to modify its fillers.Therefore,multi-performance column packing has become the focus of chromatographic research.In this work,two new monodisperse porous polymer microspheres were prepared by modifying chitin with two different compounds,and their applications in chiral drug separation and antioxidant peptide screening were discussed respectively.The specific results are as follows:(1)In this study,poly(styrene-divinylbenzene)microspheres with good dispersion were prepared by two-step seed swelling method.A new type of chromatographic stationary phase was formed by modifying 3,5-dichlorophenyl isocyanate and 3,4-dichlorophenyl isocyanate onto the surface of microspheres with diazo resin as coupling.The separation performance of the new liquid chromatographic stationary phase was studied by using benzene homologue and chlorophyllin as samples.The chiral separation ability of the new liquid chromatographic stationary phase was studied through the resolution of eight chiral compounds.This kind of chromatographic packing with non-toxic environmental protection,strong stability and high separation has a certain potential value and guiding significance for the future development of chiral stationary phase.(2)In order to further explore the application of HPLC fillers in the biomedical field,the peptide library was synthesized by Fmoc solid phase synthesis and screened by chromatography,and four polypeptide chains with antioxidant activity were successfully screened.The oxidative stress model of Hep G2 cells induced by H2O2 was established,and the protective effect of four peptide chains on oxidative damage was proved.Finally,the oil-in-water anti-oxidation sunscreen was prepared,and the skin irritation test in mice showed that the sunscreen had no irritation to the skin.In the mouse ultraviolet model,it showed excellent sunscreen and antioxidant effect.This new type of polypeptide sunscreen with environmental protection,simple operation and superior antioxidant performance has a certain potential value and guiding significance for the development and application of peptides. |
