The fruit of Rosa davurica pall.is rich in resources and widely distributed.It has excellent development prospects in treatment of cardiovascular diseases,anti-aging,hypoglycemia and so on.The extraction technology of active ingredients from fruit pulp and seeds of Rosa davurica pall.were studied,and the biological activity of the extracts was explored in vitro.Normal phase silica gel column chromatography,reversed phase C18column chromatography and thin layer chromatography were used to separate the chemical components of n-butanol extract from the fruit of Rosa davurica pall.The research results were as follows:1.The extraction technology of active ingredients from fruit pulp and seeds of Rosa davurica pall.were optimizedBased on the quality of vitamin C and the absorbance of anthocyanins in the fruit pulp of Rosa davurica pall.extracts the optimum extraction process was determined by extraction solvent,p H,acidity regulator,absorption volume,soaking balance time and ultrasonic auxiliary time.Under the optimal extraction conditions,the extraction rate of vitamin C and anthocyanins were above 95%.Vacuum drying was recommended for both vitamin C and anthocyanin fluid extract of the fruit pulp of Rosa davurica pall.The content of vitamin C in dry extract was 15.46%and the color value of anthocyanin was8.07.Using the quality of proanthocyanidin and polysaccharide of fruit seeds of Rosa davurica pall.as index,the optimal extraction process was determined by investigating extraction solvent,absorption volume,soaking equilibrium time and ultrasonic assisted time.Under the optimal extraction conditions,the extraction rates of proanthocyanidin and polysaccharide were close to 95%.After alcohol precipitation,the contents of proanthocy-anidin and polysaccharide were 14.53%and 36.13%,respectively.Based on the quality of polysaccharide,total flavonoids and total saponins in the fruit pulp of Rosa davurica pall.,the optimal extraction process was determined by investigati-ng the extraction solvent,absorption volume,soaking equilibrium time and ultrasonic assisted time.Under the optimal extraction conditions,the extraction rates of three target components were above 90%.After alcohol precipitation,the contents of polysaccharide,total flavonoids and total saponins in dry extract were 42.85%,48.45%and 12.25%,respectively.After purification,the contents of polysaccharide,total flavonoids and total saponins in dry extract were 80.41%,77.27%and 39.64%.2.The biological activity in vito of fruit pulp and seeds of Rosa davurica pall.extract were investigatedThe IC50 of inhibitionα-glucosidase of vitamin C,anthocyanins,polysaccharides,total flavonoids,total saponins in fruit pulp of Rosa davurica pall.extracts,and also proanthocyanidins and polysaccharide in fruit seeds of Rosa davurica pall.extracts were both lower than that of acarbose positive control which indicated that the fruit of Rosa davurica pall.extract may have significant hypoglycemic activity in vitro.The IC50 of DPPH·clearance rate(0.0033 mg/m L,0.0221 mg/m L)of vitamin C and anthocyanin extracts from the fruit pulp of Rosa davurica pall.was slightly higher than that of positive control vitamin C(0.0026 mg/m L).The results showed that the vitamin C and anthocyanin extracts from the fruit pulp of Rosa davurica pall.had weak scavenging ability on DPPH·.The IC50 of Nitrite removal(0.8431,2.6948,1.4709mg/m L)of polysaccharides,flavonoids and saponins in fruit pulp of Rosa davurica pall.were higher than that of vitamin C(0.5291mg/m L)which indicated that polysaccharides,flavonoids and saponins extracted from fruit pulp of Rosa davurica pall.had weak ability to remove nitrite.The IC50 of hydroxyl radical clearance(0.4484mg/m L,0.6062mg/m L)of proanthocyanidin and polysaccharide extract of the fruit seeds of Rosa davurica pall.were lower than that of vitamin C(1.4151mg/m L)which indicated that the proanthocyanidin and polysaccharide extracted from fruit seeds of Rosa davurica pall.had strong scavenging ability on hydroxyl radical.3.The chemical components of n-butanol extract from the fruit of Rosa davurica pall.were separatedUsing n-butanol extract of fruit of Rosa davurica pall.as raw material,normal phase silica gel column chromatography,reversed phase C18 column chromatography and thin layer chromatography were applied to separate the chemical components.Seven monomer compounds were obtained and five compounds were characterized by Nuclear magnetic resonance spectrometer(NMR.).They were neohesperidin dihydrochalone,quercetin-3-O-β-D-glucose-7-O-β-D-gentioside,isorhamnoside-3-O-neohesperidin,2-O-gallic hyperoside,epicatechin gallate which all were founded from Rosa davurica pall.for the first time.The other two monomers were characterized by ultra-high performance liquid chromatography and tandem quadrupole time-of-flight mass spectrometry.The structural formula of two monomer compounds may be 1,2:5,6-di-O-cyclohexylidene-3-O-methyl-α-D-glucofuran-ose,9-[(E)-4-(5-{2-[(2,4-dinitro-phenyl)-hydrazono]-ethyl}-3,4-dimethoxy-tetrahydro-pyr-an-2-yl)-3-methyl-2-enoyloxy]-nonanoic acid methyl ester.
