| Cardamine violifolia is a super selenium-rich plant with strong organic selenium conversion ability,rich in selenoproteins,selenopolysaccharides and other organic selenium compounds.As the active center of glutathione peroxidase,it has strong antioxidant effect and protects the body from oxidative damage.At present,there is little research on the application of selenium-enriched active ingredients in this plant in cosmetics with anti-aging and whitening effects.Therefore,in this thesis,the active fraction in Cardamine violifolia was extracted by ethanol reflux extraction and its antioxidant properties were evaluated by the scavenging ability of active free radicals,and its whitening properties were evaluated by measuring its inhibitory effect on tyrosinase activity.The extract was added to the skin cream to evaluate its actual effect on human skin.To provide experimental basis for the development and application of Cardamine violaceum in skin care products.The main contents of this thesis are as follows.(1)The active components in Cardamine violacea were extracted by ethanol reflux method,and its antioxidant activity was used as an evaluation index.The four factors affecting the extraction process,such as ethanol volume fraction,extraction temperature,extraction time and liquid-material ratio,were optimized by single factor experiment and response surface experiment.The optimum extraction conditions were determined as follows: ethanol volume fraction 66%,liquid-material ratio 27:1,extraction temperature 75 ℃,and extraction time 3.6 h.The DPPH radical scavenging rate was 92.63% with RSD of 0.20%.(2)In order to further investigate the difference of efficacy between selenium-rich corydalis and the common one,the selenium content in the extracts of selenium-rich Cardamine violifolia and the common one were firstly determined by XRF,and their corresponding selenium contents were 18.81 mg/L and 2.01 mg/L,and then the antioxidant ability of the two extracts was compared using DPPH,hydroxyl radical and superoxide anion radical scavenging rate as indicators.The antioxidant ability of the extracts was compared.and the inhibition rate of tyrosinase activity of the extracts were examined to compare the whitening properties of the two extracts.The effect of free radical scavenging was also determined by adding different concentrations of selenoprotein to the extract of common Cardamine violifolia.Among them,the highest scavenging rates of three free radicals were 92.12%,68.07%and 79.54% for the selenium-rich Cardamine violifolia extract,which were higher than those of 85.68%,57.06% and 61.26% for the common one.The scavenging rate of free radicals and the inhibition rate of tyrosinase were improved to different degrees by adding selenoprotein to the common Cardamine violifolia extract.The above experiments indicated that the selenium-enriched Cardamine violifolia showed more significant antioxidant effect and whitening effect than the normal samples due to its higher selenium-enriched fraction.(3)The selenium-enriched Cardamine violifolia extracts to be studied were added to the emollient cream to determine their antioxidant and whitening effects in cosmetics.The ranges of the addition amounts of different components in the formulation were determined by several single-factor tests,and the formulation scheme of the emollient cream was optimized by orthogonal experiments.The results obtained by fuzzy comprehensive evaluation method were also used to determine the optimal system for the emollient cream.The conditions of the preparation process were optimized by single-factor and orthogonal tests and combined with the results of the comprehensive scoring method to obtain the optimal process.The experimental results showed that the emollient cream was stable and did not irritate the skin.The emollient cream with the addition of selenium-rich Cardamine violifolia extract had some antioxidant and whitening effects. |
