| Ojective:Theoretical research mainly discusses the theoretical basis of traditional Chinese medicine in the treatment of Sjogren’s syndrome from the theory of lung and large intestine in the motherland medicine,and sums up the six specific treatment based on "lung intestine treatment",which provides the theoretical basis for clinical treatment The experimental study focused on the pathogenesis of Sjogong Syndrome in the treatment of Sjogren’s syndrome based on traditional Chinese medicine(Ophiopogon japonicus)can regulate the immune imbalance of Th1/Th2 and Th17/Treg in the body.In addition,by administering the mouse receptor antagonist to study whether the "pulmonary consolidation" method can play a role through the VIP pathway,Th2,Th17/Treg immunological imbalance.Method:NOD mice were randomly divided into four groups:NS+PBS group,traditional Chinese medicine+PBS(PBS),and the control group(n=10)Group,NS+VIP antagonist group,traditional Chinese medicine+VIP antagonist group.The spleen primary lymphocytes were extracted from serum,submandibular gland and spleen tissue at the age of 16 and 20 weeks respectively.The ratio of Th1,Th2,Th17 and Treg cells in spleen of mice was measured by flow cytometry.In addition,CBA and ELISA The morphological changes of the mouse submandibular gland were observed by HE staining.Real time PCR was used to detect the VIP level of the submandibular gland in mice,and then the statistical analysis was carried out.Results:Th1,2,17 and Treg cytokines in serum:IFN-γ/IL-10 ratio gradually increased compared with baseline group at 16 and 20 weeks of age,and P<0.01;Compared with the control group,the levels of IFN-γ/IL-10 in the traditional Chinese medicine group were significantly lower than those in the control group(P<0.05)(P<0.05).The ratio of IFN-γ/IL-10 in 20-week-old Chinese medicine+VIP receptor antagonist group did not change significantly.Compared with baseline group,IL-17/TGF-β ratio was significantly higher than that of baseline group at 16 weeks of age,and P<0.01;compared with 16 weeks of age,IL-17/TGF-(P<0.01).Compared with the control group,the ratio of IL-17/TGF-β in the traditional Chinese medicine group was lower than that in the control group at 16 and 20 weeks of age.Compared with the traditional Chinese medicine group,There was no significant difference in the ratio of IL-17/TGF-β between the receptor antagonist group and the control group.Th1,2,17 and Treg cells in the spleen:the ratio of Th1,2,17 and Treg cells in the spleen was significantly higher than that in the baseline group at 16 weeks of age,and the ratio of Th1/Th2 was significantly higher than that of the control group Th1/Th2 ratio was significantly decreased(P<0.05).Compared with the traditional Chinese medicine group,the ratio of Th1/Th2 in the antagonist group was significantly higher than that in the control group(P<0.01).Compared with the baseline group,the Th17/Treg ratio increased gradually(P<0.05 or P<0.01)at 16 and 20 weeks of age.Compared with the control group,the VIP antagonist group(P<0.01),but there was no significant change in the ratio of Th1/Th2 and Th17/Treg in the spleen of the mice at 20 weeks of age,but there was no significant difference between the two groups.However,compared with the control group,the proportion of Th17 in the spleen of the traditional Chinese medicine group was significantly higher than that of the control group(P<0.05 or P<0.01).Mineral DNA of submandibular gland:Compared with the baseline group,the VIP mRNA of the submandibular gland decreased gradually at 16 and 20 weeks of age(P<0.05 or P<0.01).At 16 weeks of age,compared with the control group,Compared with VIP mRNA(P<0.01),there was no significant change in VIP antagonist group.There was no significant change in VIP mRNA of submandibular gland in each group at 20 weeks of age.Submandibular gland pathology score:Compared with the baseline group,the pathological score of the submandibular gland was gradually increased at 16 and 20 weeks of gestation(P<0.05).The pathological score of submandibular gland in mice was significantly lower at 16 and 20 weeks of age in the control group(P<0.05 or P<0.01).Conclusion:Ophiopogon japonicus method can play the role of subcutaneous gland protection,positive regulation of submandibular gland VIP mRNA levels,Chinese medicine side Ophiopogon japonicus is through the VIP pathway to play a role in improving the body serum and Th1/Th2,Th17/Treg in the spleen. |
