Objective:By using the Illumina MiSeq high-throughput sequencing platform,the 16s rRNA V4 region of the intestine of the three states of the heat syndrome model,the cold syndrome model and the physiological state were used to study the effects of Radix Scrophulariacea on intestinal flora Diversity of expression of the impact of further analysis of traditional Chinese medicine Radix Scrophulariacea on the body of various metabolic and medicinal relationship.Methods:Rats were randomly divided into three groups(heat syndrome,cold syndrome,physiological state),each group of 10.The heat syndrome model was modeled with 120mg/kg/d excellent A mixture.The cold model was modeled with ice water at 0℃.The three groups of Radix Scrophulariacea were given drug in accordance with the dosage of 1.95g raw medicine/Extract for 15 consecutive days.The 16s rRNA V4 region of the intestinal samples of the three groups was determined by using Illumina Miseq as the sequencing platform.The total DNA and total RNA of the intestinal samples were analyzed.(OTUs),analyze abundance,distribution and diversity of species,and differences in species abundance,KEGG analysis and community metabolic function annotations.The effects of Radix Scrophulariacea gingkoensis on the expression and diversity of macrophages in intestinal tissue of three states were investigated.Results:1.The index of abundance of three samples of hot syndrome,cold syndrome and physiology was as follows:Compared with the normal control group,the index of heat syndrome model group increased significantly,and the index of cold syndrome model group decreased significantly,The abundance index of Radix Scrophulariaceae was also significantly decreased.Compared with model group,the group of Radix Scrophulariaceae could significantly reduce the abundance index of heat syndrome model and cold syndrome model rats.Compared with the normal control group,the index of the heat syndrome model group was significantly higher than that of the normal control group,and the index of the cold syndrome model group decreased significantly,and the diversity index of the physiological group was also significantly decreased.Compared with the group of Radix Scrophulariacea,the rats were significantly lower than those in the model group.The sparse curve indicates that the depth of the sequencing is sufficient and the number of OTUs in the three states is close to the actual situation.2.There were 2 phylums(Bacteroidetes,P<0.01,Firmicutes,P<0.01)with very significant differences in the status of the thermo-model,and there were 15 genus in the higher proportion,and the community analysis showed that,and there was a significant difference between the two groups,and there was a significant difference between the two groups.The highest proportion of the genus was 25,the community analysis showed that there were 23 groups with positive correlation,and there were 13 groups with negative correlations.There were 13 groups in the negative group.In the physiological condition,there were significant differences(Bacteroidetes,P<0.01,Firmicutes,P<0.01).There were a phylums(Acidobacteria,P<0.05).There were 15 genus;There were 23 groups with positive correlation,and there were 13 groups with negative correlation.3.The metabolic function of Scrophulariaceae in heat syndrome,cold syndrome and physiological condition showed that the intensity of pathway involved in the microbial community was Membrane transport>Carbohydrate Metabolites>Amino Acid Metabolism>Relication and repair>Energy Metabolism>Translation.Conclusions:1.In this study,we found that Radix Scrophulariacea significantly reduce the number of OTUs,the enrichment and diversity index of the intestinal flora and the distribution of species and the diversity of Alpha in the model of cold syndrome,cold syndrome and physiological state,The diversity structure of the community has a certain effect,indicating that Radix Scrophulariacea has a certain inhibitory effect on the diversity of intestinal flora in rats.2.Through the analysis of the metabolic function of the flora,the most important metabolic pathways of Radix Scrophulariacea were amino acid metabolism,glycogen metabolism and energy metabolism.Three states of the three metabolic pathways are mainly through the Firmicutes and Bacteroides diversity index and the number of changes in the impact3.Radix Scrophulariaceae on the body as a whole energy metabolism effect of inhibition,speculation that the overall performance of traditional Chinese medicine Scallion cold,but Scrophulariaceae cold medicine may contain a certain energy metabolism to promote the material basis,its performance may be with the body State has relevance.
