| Objective To investigate the mechanism of peanut red skin in treating chemotherapy-induced thrombocytopenia.Method 1.By peritoneal injection of cyclophosphamide,the model of chemotherapy-induced thrombocytopenia was successfully established.2.The successful established modle randomly divided into 6 groups:model group,low dose group with peanut red skin,middle dose group with peanut red skin,high dose group with peanut red skin,interleukin-11(IL-11)and a blank group.3.To IL-11 group,IL-1 was given by subcutaneous injection;to the peanut red skin low、medium and high dose groups,peanut red were administrated;to control group and model group,normal saline were administrated.4.The spleen of mice was weighed to calculating the spleen index.5.The expressions of caspase-7、caspase-3、Bax and Bak were detected by Western Blot.Result 1.The model of chemotherapy-induced thrombocytopenia was successfully established.2.After one week,the PLT of the peanut red skin low、medium and high dose groups and IL-11 group were significantly higher than the PLT of model group(P<0.01).3.The spleen index of the peanut red skin low、medium and high dose groups and IL-11 group were significantly higher than the spleen index of model group(P<0.01).4.The peanut red skin low、medium and high dose groups can downregulate Bax,Bak,Caspase-3 and Caspase-7 protein expression.Conclusion Peanut red skin can increase peripheral blood platelet levels,has the therapeutic effect to chemotherapy induced thrombocytopenia mice.Peanut skin reverse the spleen deficiency,protect the immune organs(spleen),improve immunity.Peanut skin downregulate the expression of Bax、Bak、Caspase-3 and Caspase-7,inhibiting platelet apoptosis. |
