Objective To investigate the influence of different dialysis time on the expression of cytokines related to the transdifferentiation of intermediate skin cells in peritoneal dialysis waste liquid,so as to provide experimental basis for the prevention and treatment of peritoneal fibrosis.Methods A total of 92 patients undergoing maintenance peritoneal dialysis in our hospital from May 2014 to April 2017 were evaluated for peritoneal dialysis quality evaluation,clinical data including name,gender,age,primary disease,dialysis,through age,complications,occurrence of peritonitis,blood pressure control and nutritional status were recorded.The patients were treated with 2.5%glucose and liquor dialysisintraperitoneus overnight for the day before the study.10ml of waste solution was collected the following morning.According to the time of dialysis peritoneal dialysis,all patients were divided into three groups:open tube group(PD<30 days)with 28 cases,PD 1-6 months group with 35 cases and PD>6 months with 29 cases.The expression of epithelial marker protein EMT in peritoneal dialysis solution(E-cadherin,Cytokeratin),abandoned mesenchymal marker protein(Fibronectin alpha,-SMA,ER)alpha/TGF-beta 1/Smad signaling pathway related protein(ER alpha,TGF-beta 1,Smad2 and Smad3)and the transport function related protein(Smad7,BMP-7)were determined using ELISA method.Result 1.With the time of dialysis,epithelial marker protein(E-Cadherin,Cytokeratin)and transport related proteins(Smad7,BMP-7)expression in peritoneal dialysis waste solution gradually decreased,which in PD>6 months group decreased more significantly;2.With the prolongation of dialysis time,the expression of fibronectin and alpha protein(-SMA)in peritoneal dialysis waste liquid increased gradually,and the decrease was more obvious in PD>6 months group;3,With the time of dialysis,the levels of ER alpha/TGF-beta 1/Smad signaling pathway related protein(ER alpha,TGF-beta 1,Smad2 and Smad3)in peritoneal dialysis liquid waste solution increased gradually,and in June,PD group increased more significantly;4.With the prolongation of dialysis time,the expression of transporters related functional proteins(Smad7 and BMP-7)decreased gradually in the peritoneal dialysis waste solution,and the decrease was more obvious in the PD>6 months group;Conclusion The cytokines related to the transdifferentiation of the intermediate skin cells in peritoneal dialysis waste may be involved in the peritoneal fibrosis process in peritoneal dialysis patients.
