Rare saponins have high pharmacological activities,this research aimed at the biological renovation and decoration of saponin structure of Panax notoginseng by biotransformation,in order to obtain novel saponins with higher pharmacological activities.The first chapter reviews the progress of chemical constituents in Panax notoginseng,pharmacological research,structural modification research and the significance of the research were summarized respectively.This article systematically screen strains with activity of biotransformation of PNS,and the conversion activity of the strains were carried out,through the steps of extraction and purification of the fermention,then obtained the converted products,eventally.Firstly,screening of active strains.Begining from the strain screening.In this paper,we screened and purified microorganisms from stereo cultivation soil of Panax notoginseng,adherent fungi of metamorphic food,endophytes of Panax notoginseng and Eupatorium adenophorum,respectively,and their conversion activity of total saponins were also tested.Finally,thirty two strains were isolated,those endophytes Zj-6(Coniochaeta sp.),Zj-8,Zj-9,Zj-10 and Zj-15 isolated from Eupatorium adenophorum have higher activity of saponin conversion.At the same time,we adopted a fast trace screening container to test the activity of strains,which can make the screening of strain with conversion activity of saponins more rapid and efficiency.Secondly,the fermentation coversion experiment of active strains was carried out.Six natural saponins of Panax notoginseng were determinated by high performance liquid chromatography,and getting six linear regression equations of notoginseng saponin R1,ginseng saponin Rg1,Re,Rb1,Rd and compound C-K respectively:Y=2.9 ×106X+12490(r2=0.9999),Y=3.1 × 106X+47852(r2=0.9995),Y=3.1 X 106X+30690(r2=0.9996),Y=2.1 × 106X+31081(r2=0.9997),Y=3.0 × 106X+37184(r2=0.9997),Y=5.7 × 106X+1.9 × 105(r2=0.9992).The saponins coverted products Rd and C-K by Zj-6 had carried on the quantitative analysis.Then the part of fermentation conditions of the strain Zj-10 and Zj-15 were optimized,at the same time the activity of crude enzymes in the fermented liquid were investigate.According to the results of biotransfromation,we choose the Zj-10 and expand the liquid shaking fermentation and fermenter tank transformation experiments.Thirdly,nine saponin constituents were obtained from fermentation pro-ducts by separ ation and purification,Quinquenoside-F1(1,Vina-ginsenoside R13(2),Notogin-senoside J(3),Pseudo-ginsenoside RT4(4),Pseudo-ginsenoside RT5(5),3-O-β-D-glu-copyranosyl-(1→2)-βD-glucopyranoside-12β,24ξ,25-trihydroxydammar-(E)-20(22)-ene(6),Vina-ginsenoside R10(7),Quinquenoside L16(8)和3β,12β,20(S),24ξ,25-O-pentahydroxy dammarane-3-O-β-D-glu copyranosyl-(2-1)-β-D-glucopyranosyl-20-O-β-D-glucopyranoside(9),respectively.Among the m,compounds 6 and 9 are new compounds.The last,three kinds of a large number of saponin were carried on pharmacological experiments and summary and outlook the biotransformation of Panax notoginseng.
