Objective To screen the effective Wuhu recipe and observe the role of Wuhu optimized formula on sugar tolerance,insulin secretion on healthy mice and diabetes mice induced by alloxan.To preliminarily study the mechanism of Wuhu optimized formula in diabetes mellitus.Methods 1.Wuhu optimized formulas were determined by orthogonal design:50 ICR mice were randomly divided into 10 groups.5 only reserved for normal group,the rest were in orthogonal design according to the L9(34)orthogonal design.Based on the fasting bloodglucose(FBG)and random blood glucose(RBG),Wuhu optimized formulas 1-1 and 1-2 were confirmed from 9 prescriptions.2.Observe the optimization of 1-1 and 1-2 on glucose tolerance in normal mice,the best one was selected:40 ICR mice were randomly divided into normal group,positive drug group,Wuhu optimized formula 1-1 and Wuhu optimized formula 1-2.Mice in normal group were gavaged with the same amount of normal saline(NS).After 10 days,mice were given oral glucose to observe the blood glucose with time(0min,30min,60min,120min).3.To observe the effect of Wuhu optimized formula on hypoglycemic effect and the mechanism in mice of alloxan-induced diabetic:randomly divided into normal group,model group,positive control group and the large,middle and small dosage of Wuhu optimized formula group.Normal group and model group were feed with equal volume of NS for 2 weeks.① FBG of mice was tested before and after dosing 2 week.②After 2 week,except for the normal group,the mice took glucose(2.5g/kg)for OGTT.The blood glucose(0min,30min,60min,120min)were measured.Insulin,SOD and MDA were also measured in the end.4.To observe the effect of Wuhu optimized formula on immune and anti-stress ability of normal mice:randomly divided into normal group,positive control group and the large,middle and small dosage of Wuhu optimized formula group.①The phagocytosis of the mice abdominal cavity macrophage was observed.②The effects of Wuhu optimized formula on the survival time of mice under anoxia were observed.Results 1.Two of Wuhu optimized formula:Yam 20g,Polygonum cuspidatum 12g,Schisandra 6g or Yam 20g,Polygonum cuspidatum 9g,Schisandra 6g.2.Wuhu optimized formula(Yam 20g,Polygonum cuspidatum 12g,Schisandra 6g)was more effective in reducing blood sugar levels.3.The large,middle and small dosage of Wuhu optimized formula group had superior hypoglycemic effect and the effect was comparable with the positive control group.In addition,Wuhu optimized formula markedly increased insulin secretion,increased the activity of SOD and reduced the content of MDA.4.The phagocytic index and the survival time of mice under anoxia in large and middle dosage of Wuhu optimized formula group increased significantly than that of normal group.Conclusion Wuhu optimized formula(Yam 20g,Polygonum cuspidatum 12g,Schisandra 6g)have good treatment effects on diabetic mice induced by alloxan though increasing insulin secretion,enhancing resistance to oxidative stress,reducing lipid peroxidation,enhancing the body immunity andimproving the ability of anti-stress.
