Objective:The aim of the study is to investigate the effect of alcohol extract of Hemerocallis citrina Baroni(HCA)on the depression-like behavior of animals and the underlying mechanisms.Methods:(1)Establish a model of simulated weightlessness by hindlimb suspension(HLS)in rats,Establish a model of chronic unpredictable mild stress(CUMS)on mice,Detect the changes in depression-like behavior by the animals behavioral of open field test(OFT),tail suspension test(TST),forced swimming test(FST)and or.(2)Investigate the antidepression effects of the HCA on the depression-like behaviour induced by the simulated weightlessness model in rats and the CUMS model in mice in rats.(3)The neurotransmitter levels(5-HT,DA,E,and HVA)in the brain of the rat after HLS were measured by the liquid chromatography-tandem mass spectrometry(LC-MS/MS).The changes of MDA,SOD,CORT,ACTH and CRF in serum of the rat after HLS were measured by biochemical analysis.Results:1.The result of the experiment of simulated weightlessness in rats:The HLS can decrease the index of source preference,the latency on the novel object test(NOT),and the moment the activity of rat in open field test.2.The result of the experiment of CUMS model in mice,it can be seen that CUMS can significantly reduce the movement duration on the central region activity of mice,increase the activity in the corner region,the latency of Novelty-suppressed feeding test(NSFT),the immobility time in TST and FST.3.The result of the HCA on simulated weightlessness model:HCA 1,2 mg/kg can decrease the source preference index,the latency of NOT and NSFT,the immobility of FST in rats,and increased the activity in the central region;Paroxetine 10 mg/kg can increase the activity of HLS rats,reduce the explore latency of NOT and NSFT,and the immobility of FST.4.The result of the HCA on CUMS model:HCA 1,2 g/kg can improve the decline of source preference index,the latency of NSFT,and the immobility of TST and FST;Paroxetine 10 mg/kg could improve the latency of NSFT and the immobility of TST.5.The result of the mechanism of the improvement effect of HCA on the simulated weightlessness model of depression-like behavior,HLS significantly decreases in E,5-HT and HVA levels.HCA 2 g/kg could significantly increase HVA levels in cortex,and significantly reduced the content of CORT,ACTH and CRF.The HCA 2 g/kg can increase the levels of these hormones;HLS can lead to increase the MDA levels and decreased SOD levels,and HCA can increase the activity of SOD while inhibited the concentration of MDA in the serum of the HLS rats.Conclusion:The HLS and the CUMS both can cause the depression-like behavior change in animals,and chronic corticosterone injection can lead to depression impairment in mice;The HCA has antidepressant effect,and we are also found that this effect may be related to the improvement of the serum tissue oxidative stress indicators MDA,SOD and the hormone levels of CORT,ACTH,CRF in rat.
