Analysis Of TCM Physique Types And Point Mutation Of ABCB4 Gene In Intrahepatic Cholestasis Of Pregnancy

Objective To investigate the TCM physique type and point mutation of ABCB4 gene in intrahepatic cholestasis of pregnancy,and provide the basis and reference for the development of targeted treatment for intrahepatic cholestasis of clinical pregnancy.(1)Methods Experimental methods:150 cases of ICP patients admitted to the department(2)between September 2017 and February 2018 were selected and divided into blood deficiency type group and hepatobiliary and dampness type group according to TCM classification.Among them,30 cases were selected for the each traditional Chinese experiment group,such as Qi-qi stagnation group,qi-deficiency group,phlegm-dampness group,blood stasis group and ping-qi group,At the same time,30 normal pregnant women of the same period of middle and late pregnancy were selected as the control group.DNA samples of 150 specimens of peripheral blood were amplified by PCR,and the PCR products were determined by DNA sequencing.The serum samples were separated by DNA,DNA extracted and ABCB4 genotypes were determined.And healthy controls in patients with serum ABCB4 gene mutations in three high-risk sites 6,8,14 exon mutations.(2)Mathematical Statistics:SPSS 18.0 software was used to process count data line χ2 test,using n(%),the test standard a is 0.05,P<0.05 represents the difference Statistical significance.Results The DNA fragments of exon 6,8 and 14 of ABCB4 gene were amplified from DNA samples of 150 patients with ICP and 30 normal controls,and the target fragments were amplified.In addition,there was no significant difference between PC R-PCR and DNA sequencing in patients with control group and experiment group(P>0.05).The results also showed that ABCB4 gene exon 8 and exon Sub-14 abnor-mal.The results showed that the base 504 of ABCB4 gene exon 6 was changed in 48 cas-es of ICP patients,among which 23 cases were c.504C>T homozygous mutat ion and 25 cases were c.504C>T heterozygous mutation.Conclusions Synonymous mutations in exon 6 of ABCB4 gene may be associated with the occurrence of ICP in Chinese population.Exon 8 and 14 can not be confir med temporarily with ICP in Chinese population.Hlegm-dampness group and blood stasis group may affected the ABCB4 gene...
