Objective :By a high-fat diet,build C57 mice model of nonalcoholic fatty liver disease,in order to verify the miR-22 change in nonalcoholic fatty liver disease and function,discusses whether it is associated with FXR,SIRT1,FGF21 related pathways.Methods:16 males to 8 weeks of C57 mice,adaptive feed after 1 week,both two groups at random,blank control group and high-fat diet group,respectively,respectively,to the ordinary feed and high fat diet for 8 weeks,the experimental observation in the process of animal growth,behavior,and shit.9 weeks after the experiment,unified treatment was performed: 1.Blood collection was collected for biochemical index detection;2.Dissect mice and extract liver tissue for observation;3.The expression of FXR,SIRT1,FGF21 and mir-22 m RNA were detected using qrt-pcr,and the protein content of FXR,SIRT1 and FGF21 were detected by Western Blotting.Result:1.At the end of the 9th week,mice fed with high-fat diet had lower body weight than those in the blank control group(P<0.05).The liver weight was significantly higher than that in the blank control group(P< 0.05),and the liver index was significantly higher than the blank control group(P< 0.05).2.Serum biochemistry suggested that the total cholesterol(7.39 ± 2.42),low-density lipoprotein(1.94 ± 0.89),aspartate aminotransferase(246.17 ± 61.86),and alanine aminotransferase(424.86±220.39)related indexes were higher than the blank control in the high-fat diet group.Total cholesterol(2.29±0.64),low-density lipoprotein(0.26 ± 0.18),aspartate aminotransferase(128.6 ± 32.55),and alanine aminotransferase(39.00±9.30)increased significantly(P<0.05).3.The expression of FXR and SIRT1 m RNA in NAFLD model group was significantly lower than that in normal group,but the expression of FGF21 m RNA was significantly higher than that in normal group(P<0.01).The protein levels of FXR,SIRT1 and FGF21 in model group were significantly higher than those in normal group.The expression levels were significantly decreased compared with the normal group,and the difference was statistically significant(P<0.05).The expression of miR-22 in the NAFLD model group was significantly higher than that in the normal group(P<0.05).Conclusion1.In the NAFLD model,the m RNA and protein levels of the FXR and SIRT1 genes were significantly reduced,and the FGF21 gene m RNA level was significantly increased,while the protein level was significantly reduced.2.In the NAFLD model,miR-22 expression was significantly increased compared with the normal group.FXR and FGF-21 are the target genes of miR-22 at the same time.It does not exclude the existence of miR-22-related pathways regulating FXR and FGF21,which may be related to the formation of NAFLD.
