Objective:To extract and separate the chemical components of Potentilla longifolia,and identify the chemical structures of the obtained monomer compounds and test their inhibitory effects on lipid accumulation in 3T3-L1 preadipocytes.Methods:The aerial part of Potentilla longifolia was extracted by 95%ethanol to extract the reflux and obtain the crude extracts.Then,the root of Potentilla longifolia was decocted with distilled water to obtain the crude extracts of the root of Potentilla longifolia.The crude extracts were purified by different separation purification methods(Silica gel column chromatography,Octadecylsilyl column chromatography,Macroporous adsorption resin column chromatography,MPLC,HPLC and Sephadex LH-20).And the structures of compounds were identified by various nuclear magnetic resonance means(1H-NMR,13C-NMR,HMQC,HMBC).Conclusion:It contains ten known compounds and one new compound.These structures were indentified as 1-(6’-hydroxy-4’-methoxyphenyl-2’-O-β-D-glucopyranoside)-3-methylbutanone(1),8-O-methylherbacetin-3-O-β-D-sophoroside(2),Quercetin-3-O-(2"-O-β-D-glucopyranosyl)-β-D-xylopyranoside(3),Quercetin-3-O-β-D-glucopyranoside-(3’→O-3’")-quercetin-3-O-β-D-galactopyranoside(4),Quercetin-3-O-β-D-glu-copyranoside(5),(+)-pinoresinol(6),3β,19α-dihydroxy-1-oxo-urs-12-en-28-oic-acid(7),Quercetin-3-O-β-D-galactoside-7-O-β-D-glucoside(8),(+)-catechin(9),tianshic acid(10),1-(3’-methyl-4’-methoxyphenyl-2’,6’-di-O-β-D-glucopyranoside)-2-methylbutanone(11).Among them,compounds 3,4,6,11 were separated from the Rosaceae plant for the first time.Compound 2,3,4,7,11 were isolated from aerial parts of Potentilla longifolia for the first time.Compound 6,8,9,10 were isolated from the root of Potentilla longifolia for the first time.Compound 11 is a new compound.In addition,the compounds isolated from Potentilla longifolia were used to screen the active components of 3T3-L1 preadipocytes.The results showed that compound 5,11 extracted from Potentilla longifolia had a good inhibitory effect on 3T3-L1 preadipocyte differentiation and lipid accumulation.
