| Empetrum nigrum L.var.japonicum K.Koch is the only variant of Empetraceae in China.It is mainly distributed in the Greater Khingan Mountains in northeast China;it grows in stone mountains or forests at an altitude of about 1200-1400 m,and it has the characteristics of resistance to cold,drought,barren,light,and wind.Ancient folk records that the E.nigrum var.japonicum whole grass can be used as a tea for health,with the effect of.nourishing and storing energy.It can be used as a medicine,branches and leaves can strong the spleen and stomach,can treat abdominal fullness,and indigestion diseases.Leaves and roots have diuretic and nourishing effects.The fruit nourishes the yin and the liver,it can be used to treat hepatitis,heart disease and other diseases in the folk.It has a sweet and sour taste,and can be eaten raw.It can be used to make wine,but also to make jams,jellies,drinks,and extract food coloring.The medicinal value of E.nigrum var.japonicum is very high.In order to explore its effective components,this experiment used methanol reflux extraction of the herbs of E.nigrum var.japonicum to obtain the methanol extract.We extracted it with petroleum ether,ethyl acetate and n-butanol.The normal-phase silica gel column chromatography,reversed-phase silica gel column chromatography,medium-pressure preparative chromatography(ODS),and dextran gel chromatography(Sephadex LH-20)were used to separate from the methanol extraction of E.nigrum var.japonicum.A total of 12 monomer compounds were obtained.According to the physical and chemical properties of the monomer compounds,spectral data analysis and reference literature comparison,the above identified monomer compounds were structurally identified,and their structures were finally determined.These compounds were identified as 4’-hydroxy-2’,6’-dimethoxy-3’,5’-dimethyl-dihydrochalcone(1),2’,4’-dihydroxy-dihydrochalcone(2),2’,4’-dihydroxy-chalcone(3),β-sitosterol(4),2’,5’-dihydroxy-3’-methoxy-4’,6’-dimethyl-dihydrochalcone(5),2’-methoxy-4’-hydroxyl-α,β-dihydrochalcone(6),2’,4’,β-trihydroxy-dihydrochalcone(7),11α-methoxyurs-12-ene-3β,12-diol(8),Davidigenin(9),(3-(4-hydroxyphenyl)-1-propanol(10),Ursolic acid(11),Uvaol(12).Compound 1 is a new compound found in E.nigrum var.japonicum.Compound 8,9 and 10 was isolated from E.nigrum var.japonicum for the first time.After reviewing the literature,we found that the flavonoids and the triterpenoids are the main components of E.nigrum var.japonicum.Determination the content of compound 2,3 and 6 by the HPLC method.The results show that compound 2 showed a good linear relationship in the range of 5.000~0.078 mg/mL(r=0.9998),compound 3 showed a good linear relationship in the range of 5.000~0.078 mg/mL(r=1.000),compound 6 showed a good linear relationship in the range of 4.000~0.063 mg/mL(r=1.000).It can be seen that this method is simple,convenient,reliable,and reproducible,which can provide experimental basis for the future formulation of quality standards of E.nigrum var.japonicum medicinal materials and extracts. |
