Polygonati Rhizoma is a perennial herbaceous plant in the Polygonatum Mill.family,and it is a medicinal and edible plant.At present,the pharmacopoeia stipulates that the three medicinal Polygonati Rhizoma are Polygonatum cyrtonema Hua,Polygonatum kingianum Coll.et Hemsl and Polygonatum sibiricum Red.Among them,the source of Jiuhua Polygonati Rhizoma in Anhui Province is P.cyrtonema Hua,which is one of the "Top Ten Anhui Medicines".In order to understand the reasons for the famous and famous Jiuhua Polygonati Rhizoma,we compared the quality of three medicinal Polygonati Rhizoma.In this paper,the content of tuber polysaccharides in different growth years of three kinds of Polygonati Rhizoma was tested,and the reducing power of the polysaccharides of Polygonati Rhizoma and its ability to remove DPPH and ABTS were studied;use GC-MS technology to extract and detect metabolites of three kinds of Polygonati Rhizoma tubers and P.cyrtonema Hua with different growth years,combined with multiple statistical methods,screening differential metabolites of three kinds of Polygonati Rhizoma,in order to find out the metabolites that affect the antioxidant capacity of Polygonati Rhizoma polysaccharides,and provide a theoretical basis for the development and utilization of Polygonati Rhizoma.The main experimental results are as follows:1.The polysaccharides in the three kinds of Polygonati Rhizoma tubers were significantly different,and they were P.cyrtonema Hua> P.kingianum Coll.et Hemsl> P.sibiricum Red.The total amino acid content is P.kingianum Coll.et Hemsl> P.sibiricum Red> P.cyrtonema Hua.Among them,the polysaccharide content of P.cyrtonema Hua tuber is: third year> second annual> first year;the polysaccharide content of P.sibiricum Red tuber is: second year> first year> third year;the polysaccharide content of P.kingianum Coll.et Hemsl tuber is: third year>second year> first year.It shows that the quality of P.cyrtonema Hua is better than that of P.sibiricum Red and P.kingianum Coll.et Hemsl.2.The tuber extracts of Polygonati Rhizoma have certain removal ability to DPPH and ABTS.The overall performance is that P.cyrtonema Hua is significantly better than P.kingianum Coll.et Hemsl and P.sibiricum Red(p <0.05).Polygonati Rhizoma has a strong ability to scavenge DPPH and ABTS free radicals.The antioxidant performance of the three Polygonati Rhizoma overall performance is that P.cyrtonema Hua is significantly better than P.kingianum Coll.et Hemsl and P.sibiricum Red(p <0.05).3.Based on gas chromatography and mass spectrometry(GC-MS),the preparation and detection methods of methanol extract of Polygonati Rhizoma sample are optimized.Extraction method: ultrasonic 30 min,oxime time 120 min,oxime temperature 60℃,oxime The reagent(hydroxylamine hydrochloride)dosage is 60 μL,the derivatization time is 90 min,the derivatization temperature is 60℃,and the dosage of the derivatization reagent(99% BSTFA + 1% TMCS)is 60 μL.Detection method: split ratio 10: 1,heating program: 8℃/min,solvent delay time 14 min.The methanol/water phase extracts of the tuber sample of Polygonati Rhizoma are mainly sugars and glycosides,amino acids,fatty acids,organic acids,alcohols and esters,alkanes,etc.4.Using GC-MS technology,92,65 and 69 metabolites were identified from the tuber of P.cyrtonema Hua,P.sibiricum Red and P.kingianum Coll.et Hemsl,respectively.Alloxanose,erythrose,ribose,etc.have the highest content in P.kingianum Coll.et Hemsl,trehalose,sucrose and other sugars and glycosides have the highest content in P.cyrtonema Hua;ribofuranose,D-xylose,triose Reducing sugars were only detected in P.cyrtonema Hua.13 differential metabolites were selected from P.sibiricum Red and P.kingianum Coll.et Hemsl;19 differential metabolites were selected from P.sibiricum Red and P.cyrtonema Hua;17 differential metabolites were selected from P.kingianum Coll.et Hemsl and P.cyrtonema Hua.The content of ribose in P.kingianum Coll.et Hemsl and P.sibiricum Red is higher than that of P.cyrtonema Hua.Both reducing sugars,fructose and glucose,have the highest content in P.cyrtonema Hua.5.Fourteen different metabolites were selected from the first year and second year tuber metabolites,of which two kinds of sugar and glycosides were trehalose and cellobiose;four differential metabolites were screened from the second and third year tubers,of which two were sugar and glycosides,trehalose and cellobiose,respectively.The sugars and glycosides in the tuber of P.cyrtonema Hua are the third year>the second year>the first year. |
