Background:China is one of the regions with high incidence ofesophageal squamous cell carcinoma(ESCC).Duoto lack of early detection and targeted drugs for ESCC,the incidence and mortality of ESCC still remain high in China for a long time.Therefore,it is very important to study the molecular mechanism and target genes of ESCC.In recent years,the rapid development of bioinformatics has provided new idea forstudyingESCC.Objective:This study aimed to investigate the RAD21 expression in ESCC,and analyze the correlation of RAD21 with the clinico-pathological characteristics and prognosis,then explored the potential clinical significance of RAD21 in ESCC.Methods:1.Analyze the different genes of GSE20347 data and screen forthe core gene RAD21 by using Cytoscape software.2.The expression of RAD21 in ESCC was analyzed in Oncome database.3.RNA and protein were collected from ESCC and its adjacent tissues from patients,whounderwent esophagectomy at the Department of Cardiothoracic Surgeryin Affiliated Hospital of North Sichuan Medical College from November2018 to May 2019.RT-qPCR and Western blot were used to detect the expression of RAD21 in ESCC.4.Using TCGA database,81 patients with clinical information about each axial squamous cell carcinoma were selected to compare the correlation between the expression level of RAD21 mRNA and the clinicopathological characteristics and the prognosis of ESCC.5.GSEA Software 4.0 was used to analyze the potential biological function of RAD21 in TCGA database.Results:1.Compared with the adjcent tissues,RAD21 expression of ESCC was up-regulated in mRNA and protein levels.2.Meta-analysis of oncome data showed that the expression of RAD21 was significantly up-regulated in multiple researchcenters aboutESCC.3.RAD21 expression was closely related to tumor size(P=0.029).4.TCGA database and Kaplan Meier analysis showed that the high expression of RAD21 was significantly related to the good prognosis.5.Multivariate regression analysis showed that RAD21 could be used as an independent prognostic indicator of ESCC(HR=0.95/P=0.006).6.GSEA analysis showed that RAD21 was closely related to cell cyclesregulation and homologous recombination gene set of chromosome.Conclusion:RAD21 is highly expressed in ESCC,and may be an important prognostic indicator.GSEA analysis of RAD21 may be involved in the genesis and development of ESCC by regulating cell cycle,homologous recombination of chromosomes.
