Cannabidiol(cannabidiol,CBD)is a cannabinoid in hemp.Modern medicine shows that it has a wide range of pharmacological effects,such as anti-epilepsy,anti-inflammation and antioxidation,cardiovascular protection and so on,but its application is greatly limited because of its bioavailability.Nanoemulsion is a new type of carrier with great research potential,with smaller particle size of emulsion droplets compared to ordinary emulsions,which can improve the bioavailability of drugs,and with the development of new drugs,nanotechnology can also increase the water solubility of drugs,and nanoemulsion has a wide range of applications as a carrier system in the fields of medicine,food,and biotechnology,chemical-like technology,environmental science,etc..In this paper,cannabidiol(CBD)was used as raw material to select the formula of cannabis diphenol nanoemulsion,and the preparaton technique was optimised by single factor experimental design and response surface optimization.Based on this,the physical and chemical properties and stability analysis were carried out,and the in vitro digestion experiments and antioxidant properties were studied,which provided a theoretical basis for the application of cannabis diphenol nanoemulsion in the field of medicine,food and health products.The main research contents are as follows:The main results are as follows:(1)The selection of each phase of the nanoemulsion formulation was carried out to screen the oil phase from six different edible oils,and the oil phase was selected as corn oil by the solubility of CBD in edible oil;the complex surfactant with HLB=12 was selected by the pseudo-ternary phase diagram,anhydrous ethanol was selected as the co-surfactant,and the most suitable mass ratio of surfactant to co-surfactant was Km=2:1.(2)The formula and process preparation were optimized.Through response surface design,the best formula of CBD nano-emulsion was obtained as follows:corn oil 1.20g,distilled water 6.02g,mixture of surfactant and co-surfactant 5.61g.The preparation process of nano-emulsion was optimized by single factor experiment and response surface design,and the best optimal technological condition as follows:stirring time was 17 minutes and water phase temperature was 27℃.When the drop acceleration is 1.0 mL/min,the dosage is 45.5mg.(3)Through the physical and chemical characterization and stability experiments of the prepared CBD nanoemulsion,it is found that CBD can improve the viscosity of the nanoemulsion,and the water solubility of CBD nanoemulsion is very good.And when the room temperature and air temperature change,the particle size of nano-emulsion does not change much,it is not layered for a long time,the light sensitivity is weak,and basically maintain the original particle size and appearance.(4)Through the digestion experiment of simulated CBD powder and nano-emulsion in oral fluid,gastric juice and intestinal juice,through the determination of the content of CBD,it was found that nano-emulsion could protect the effective substances from being destroyed and could be absorbed into intestinal fluid.Through the dialysis bag,the bioavailability was improved.(5)Using freeze-drying method,CBD nanoemulsion was cured into lyophilized powder,and the physical and chemical properties were tested,and it was found that the nanoemulsion lyophilized powder formulation could well encapsulate the active substance and protect the CBD active substance to reduce external damage,thus improving the stability.(6)The prepared CBD nanoemulsion and lyophilized powder were subjected to DPPH and ABTS+in vitro antioxidant capacity assays,and Vc was used as a control for the analysis of antioxidant capacity.From the above two in vitro antioxidant assays and IC50 value calculations,it was found that both CBD nanoemulsion and nanoemulsion lyophilized powder had good antioxidant activity and the strength of antioxidant capacity were both:Vc>CBD nanoemulsion lyophilized powder>CBD nanoemulsion,indicating that CBD nanoemulsion prepared as lyophilized powder had enhanced antioxidant capacity.This is because CBD nanoemulsion lyophilized powder is more soluble in water,easy to react with DPPH and ABTS+,and β-cyclodextrin also has a good antioxidant capacity.To sum up,the nano-emulsion prepared by optimized formula and optimized process is more stable than CBD powder,and the ability of digestion and antioxidation in vitro has been improved.Therefore,nano-emulsion preparation can be used as the future development direction of CBD raw materials,and also provide some data support for further focusing on CBD processing technology and developing new CBD products in the future.
