Evaluation Of MORC Family Genes On Prognostic Survival Of Gastric Cancer Patients

Objective To investigate the expression levels of MORC family genes(MORC1,MORC2,MORC3 and MORC4)on the prognostic survival(Overall survival(OS),Progression Free Survival(PFS),Post Progression Survival(PPS))of gastric cancer patients.Methods This project was conducted by accessing the online Kaplan-Meier Plotter database(http://kmplot.com/analysis/).This database has survival information of 881 gastric cancer patients.In this study,we used Kaplan-Meier Plotter analysis to explore the relationship between the expression levels of the MORC family(MORC1,MORC2,MORC3,MORC4)and prognostic survival in gastric cancer patients.The expression of MORC family genes was classified into"low" and "high" expression according to the median levels of gene expression in gastric cancer samples,and the relationship between the mRNA expression levels of genes and prognostic survival time(OS,PFS,PPS)of patients was counted.Among them,MORC family genes had OS in a total of 875 cases,PFS in a total of 640 cases,and PPS in a total of 498 cases.In addition,we analyzed the relationship between MORC family members and prognostic relevance and clinical characteristics of patients(TNM stage,HER2 expression and different treatments).Results The mRNA expression levels of MORC 1 were negatively correlated with OS in 875 gastric cancer patients(HR=1.58(1.31-1.91),P<0.0001),i.e.,the higher the mRNA expression levels of MORC 1,the worse the OS and PFS of gastric cancer patients;it was also negatively correlated with PFS in 640 gastric cancer patients(HR=1.42(1.16-1.74),P<0.001);while there was no correlation with PPS in 498 gastric cancer patients(HR=1.24(0.99-1.54),P>0.05).The expression levels of mRNA of MORC2 were negatively correlated with OS in 875 gastric cancer patients(HR=1.73(1.46-2.05),P<0.0001);it was also negatively correlated with PFS in 640 gastric cancer patients(HR=1.92(1.572.35),P<0.0001);and it was also negatively correlated with PPS in 498 gastric cancer patients(HR=1 2.02(1.6-2.54),P<0.0001).The mRNA expression levels of MORC3 positively correlated with OS in 875 gastric cancer patients(HR=0.52(0.43-0.62),P<0.0001),i.e.,the higher the mRNA expression level of MORC3,the better the OS of gastric cancer patients and vice versa;it was positively correlated with the PFS of 498 gastric cancer patients(HR=0.52(0.43-0.64),P<0.0001).Positive correlation was found with PFS in 640 gastric cancer patients(HR=0.5(0.4-0.63),P<0.0001).The mRNA expression levels of MORC4 were positively correlated with OS in 875 gastric cancer patients(HR=0.8(0.66-0.97),P<0.05),but not with PFS and PPS in 640 gastric cancer patients(HR=0.88(0.71-1.1),P>0.05;HR=1.2(0.94-1.55),P>0.05).The different MORC1 expression levels were correlated with OS in patients with stage Ⅰ and Ⅲ TNM stage,with statistical differences(P<0.05);the different expression levels of MORC2 and MORC3 were correlated with OS in patients with stage Ⅰ,Ⅱ and Ⅲ gastric cancer(P<0.05);and the different expression levels of MORC4 were correlated with OS in patients with stage Ⅳgastric cancer.The different expression levels of MORC family were significantly correlated with OS in patients with different HER2 expression(P<0.05).The different expression levels of MORC 1,MORC3 and MORC4 were statistically different in the 5-Fu-based adjuvant chemotherapy and adjuvant chemotherapy with other drugs groups(P<0.05),but not in the surgery-only group(P>0.05);MORC2 was statistically different in the surgeryonly and 5-Fu-based adjuvant chemotherapy groups(P<0.05),while there was no statistical difference in adjuvant chemotherapy with other drugs(P>0.05).Conclusion The expression levels of MORC family genes have a predictive effect on the prognostic survival time of gastric cancer patients,and the expression levels of MORC1 and MORC2 were negatively correlated with the prognostic survival time of gastric cancer patients;while the expression levels of MORC3 and MORC4 were positively correlated with the prognostic survival time of gastric cancer patients.In addition,the expression levels of MORC family genes may have a predictive role for OS in TNM staging and different treatments.
