Objective: Inflammatory bowel disease(IBD)mainly includes ulcerative colitis(UC)and Crohn’s disease(CD).The etiology is not yet clear.This study is to investigate the correlation between Glutathione S-transferase pi(GSTP1)gene rs1695 locus polymorphism and the susceptibility and clinical characteristics of IBD in Guangxi Zhuang population and Han population,and the relationship with adverse reactions of treating IBD with Azathioprine(AZA).Methods: We selected 81 Han population IBD patients(45 UC,36 CD)and63 Zhuang population IBD patients(34 UC,29 CD)unrelated in Guangxi Zhuang Autonomous Region,and 101 healthy people(60 Han population and 41 Zhuang population).The intestinal mucosa tissue was collected by endoscopic biopsy forceps,and extracted the DNA to PCR and sequenced.Calculating and analyzing the correlation between genotype and allele frequency of GSTP1 gene rs1695 locus and the susceptibility and clinical characteristics of IBD.Meanwhile,IBD patients treated with AZA were followed up to observe the correlation between the adverse reactions and the genotype of rs1695 locus.Results: The frequencies of GSTP1 gene rs1695 locus genotypes and alleles in Han UC group,Zhuang UC group,Han CD group,and Zhuang CD group were no statistical difference from those in the normal control group(P > 0.025).No significantly difference between the genotypes and alleles frequencies of GSTP1 gene rs1695 locus in Han UC group and Han CD group were respectively discovered in those in Zhuang UC group and Zhuang CD group(P > 0.05).In Han UC group and Zhuang UC group in different lesion sites and disease activity,there was no significant difference in genotypes and alleles frequencies distribution of GSTP1 gene rs1695 locus(P > 0.05).During a12-month follow-up of IBD patients treated with AZA at the first time,no association was found between the frequency of mutations at this locus and leukopenia during AZA treatment.Conclusion: The polymorphism of GSTP1 gene rs1695 locus may not be related to the susceptibility of IBD in Guangxi Han population and Zhuang population,and has nothing to do with the lesion site and disease activity degree of UC in Han population and Zhuang population.It has not been found that the mutation genotype of GSTP1 rs1695 locus is associated with leukopenia during AZA treatment of IBD.
