| Objective: The study is to observe the effect of guava leaf extract on body weight,blood lipid and Browning of white fat in high fat induced obese C57 mice,and to explore the mechanism of guava leaf extract on weight loss mechanism in high fat induced obese C57 mice.The results will provide experimental basis for clinical application of guava leaf.Methods: Five-week-old male C57BL/6J mice were fed for 2 weeks and randomly divided into 2 groups: normal group(NC group)and high fat group(HFD group).After that,the mice in NC group and HFD group were fed with normal diet and high-fat diet respectively.During the 10 weeks of modeling,the body weight of the HFD group increased by more than 20%(including 20%)compared with that of the NC group,that is,the obesity model mice were successfully established.The HFD group was then randomly divided into four subgroups(obesity model group,namely O group,liraglutide treatment group,namely O+L group,guava treatment group,namely O+HGL group,and guava treatment group,namely O+LGL group.O+HGL group was given intragastric gavage at 1632mg/Kg,O+LGL group was given intragastric gavage at 816mg/Kg,O+L group was given subcutaneous injection at 0.2mg/K,NC group and O group were given intragastric gavage with the same volume of normal saline for 8weeks.During treatment,the body weight and body length of the mice were monitored every 4 weeks and Lee’s index was calculated.After 8 weeks of treatment,serum lipid levels and HE staining of peri-testicular adipose tissue were detected.The protein and gene expressions of UCP1,PPARγ and PGC-1α in peritesticular adipose tissue were detected by Western blot and quantitative real-time PCR.Results:1.After 8 weeks of treatment,compared with the NC group,the weight of mice in group O increased significantly(P < 0.05).Compared with the O group,the weight of mice in O+L group and O+HGL group decreased significantly(P < 0.05).Compared with O+Lgroup,the weight of O+HGL group and O+LGL group increased significantly(P < 0.05).2.After 8 weeks of treatment,compared with the NC group,the Lee’s index of mice in group O increased significantly(P < 0.05).Compared with the O group,the Lee’s index of O+ Lgroup,O+HGL group and O+LGL group decreased significantly(P < 0.05).Compared with the O+L group,the Lee’s index of O+HGL group and O + LGL group increased significantly(P < 0.05).However,there was no significant difference in Lee’s index between O+HGL group and O+LGL group.3.After 8 weeks of treatment,compared with the NC group,the levels of TG,TC,HDL-C and LDL-C in group O were all increased significantly(P < 0.05),Compared with the O group,the levels of TG,TC,HDL-C and LDL-C in O+L group and O+HGL group were decreased,with statistical significance(P <0.05).Compared with the O group,the TG level of the O+LGL group was decreased significantly(P < 0.05),but the TC,HDL-C and LDL-C levels were not significantly different.Compared with the O+L group,the levels of TG in O+HGL was increased significantly(P < 0.05),but the levels of TC,HDL-C and LDL-C were not significantly different.Compared with the O+L group,the levels of TC and TG in O+LGL was increased significantly(P < 0.05),but the levels of HDL-C and LDL-C were not significantly different.Compared with the O+HGL group,TC level in O+LGL group was increased significantly(P < 0.05),but TG,HDL-C and LDL-C levels were not significantly different.4.After 8 weeks of treatment,compared with the NC group,the abdominal fat coefficient of group O was increased significantly(P < 0.05).Compared with the O group,the abdominal fat coefficient of O+L group,O+HGL group and O+LGL group was decreased significantly(P<0.05).5.HE staining results of adipose tissue: after 8 weeks of treatment,the fat cells in NC group were small,fat vacuoles were small,the nucleus was obvious,the shape was relatively round,and the surrounding tissue was relatively dense.Compared with the NC group,the adipocytes in the O group were large,the cell contents were squeezed to one side and presented a thin ring,and the nucleus was not obvious and the shape was irregular Compared with the O group,the adipose tissue cells around the testis in the O+L group,O+HGL group and O+LGL group became smaller and rounded,with multiple locules,more obvious nuclei and denser tissues.Compared with the O+L group,the fat compartment vacuoles around the testis in the O+HGL and O+LGL groups were slightly larger,the nuclei were not obvious,and the tissue was relatively sparse.Compared with the O+HGL group,the fat compartment vacuoles around the testis in the O+LGL group were slightly larger,the nuclei were not obvious,and the tissue was relatively sparse.6.Compared with the O group,the protein and gene expression levels of UCP1,PPARγ and PGC-1α in O+L group and O+HGL group were increased significantly(P<0.05),while only the protein expression levels of UCP1,PGC-1α and gene expression levels of UCP1 in O+LGL group were increased significantly(P<0.05).Conclusion:1.Guava leaf extract can significantly delay the weight gain of C57BL/6J mice induced by high fat.2.Guava leaf extract can improve the lipid disorder and reduce the blood lipid in C57BL/6J mice induced by high fat.3.Guava leaf extract can effectively control the accumulation of abdominal fat in obese mice,and thus play a role in delaying weight gain.4.Guava leaf extract may increase body heat production and energy consumption by regulating the Browning of white fat,thus playing the role of weight reduction and lipid reduction and improving metabolism of obese body. |
