| Objective: This study aims to identify the expression profile of lnc RNA in the plasma exosomes of patients with early-onset severe preeclampsia,in an attempt to explore the molecular markers of preeclampsia in peripheral blood,and to explore the pathogenesis of preeclampsia at the molecular level.Methods: In the previous study,we extracted RNA from plasma exosomes of 5 patients with early-onset severe preeclampsia(PE group)and 5 normal pregnant women(NC group),and the top 7 lnc RNAs with strong correlation with the onset of preeclampsia were screened by bioinformatics analysis:ENST00000559730.1 、 ENST00000545188.1 、 ENST00000428958.1 、ENST00000535614.1 、 ENST00000573220.1 、 ENST00000566645.1 、 and ENST00000577807.1.The lnc RNAs transcript numbers and gene names are detailed in Appendix 1.The top 10 m RNAs significantly differentially expressed between the two groups were: B2M、PPBP、SMN2、CD99、PNMA6A、VAMP7、PGA4、FTH1、OST4 and NUDT16,as detailed in Appendix 2.In this study,we collected the peripheral blood of 5 patients with early-onset severe preeclampsia(PE group)and 5 normal pregnant women(NC group),extracted RNA from plasma exosomes,and used q RT-PCR to verify the top 7 lnc RNAs with strong correlation.The key lnc RNA,mi RNAs and m RNAs were screened by network quantitative relationship,and Pearson correlation coefficient and P value among multiple genes were calculated to construct the lnc RNA-mi RNA-m RNA co-expression network.Results: The results of candidate lnc RNAs q RT-PCR verification showed that 7 lnc RNAs were strongly correlated with the incidence of preeclampsia:ENST00000559730.1 、 ENST00000545188.1 、 ENST00000428958.1 、ENST00000535614.1 、 ENST00000573220.1 、 ENST00000566645.1 、ENST00000577807.1 were expressed in plasma exosomes of pregnant women in both PE group and NC group.Among them,the expressions of ENST00000559730.1,ENST00000545188.1,ENST00000428958.1 and ENST00000573220.1 in PE group were significantly higher than those in NC group.Lnc RNA-mi RNA-m RNA co-expression network analysis showed that ENST0000559730.1-hsa-mi R-661-NUDT16 had the highest correlation.Conclusion: There is a significant difference in the expression of lnc RNAs in plasma exosomes between patients with early-onset severe preeclampsia and normal pregnant women,suggesting that lnc RNAs may be involved in the pathogenesis of preeclampsia.Combined with our previous and this study results,we speculate that ENST0000559730.1,hsa-mi R-661 and NUDT16 may affect the development of preeclampsia through a variety of signal transduction pathways. |
