Clinical Prescription Rules Of Traditional Chinese Medicine For Adenomyosis Based On Complex Network Analysis And Its Network Pharmacology Research

Objective:Based on the real world research,the prescription data of clinical TCM for adenomyosis were collected,and various data analysis methods were used to summarize the high-frequency drug use,core drug pairs,core prescriptions and generic prescriptions of TCM for adenomyosis,providing new ideas for clinical drug use.The mined core prescriptions are studied by network pharmacology,and the drug-target-gene-disease relationship network is constructed.By analyzing the mechanism of core prescriptions from different angles,a new direction is provided for laboratory research.Methods:This study is divided into two parts.The first part is data mining and analysis.The information of patients who visited the department of Gynecology of The Affiliated Hospital of Shandong University of Traditional Chinese Medicine from January 1,2015 to December 31,2020 was extracted,and the treatment information of patients with adenomyosis that met the criteria of receiving and discharging was included in the database.Frequency analysis,Liquorice system and Gephi software were used to excavate and analyze the medication rule of adenomyosis from multiple angles.The second part is network pharmacology study.Using TCMSP database(http://tcmspw.com/tcmsp.php)to complicated network analysis is the core of the prescription medicine in compound component retrieval and target forecast,in Uniprot database(https://www.uniprot.org/)will be converted to human genes.Such as Genecards database(https://www.genecards.org/)to retrieve the womb gland myopathy associated genes.Take Chinese traditional medicine to predict target genes associated with uterine adenomyosis intersection and use its online mapping web site(http://bioinformatics.psb.ugent.be/webtools/Venn/)to make the intersection Venn diagram.The protein-protein interaction network diagram was made by String database(https://string-db.org/),and the protein functional plates were divided by the MCODE plug-in of Catoscape software.To analyze gene enrichment using Metascape database(https://metascape.org/),the results of the analysis using the microscopic letter online mapping web site(http://www.bioinformatics.com.cn/)for visual analysis.Results:1.A total of 5791 prescriptions were collected,including 347 kinds of Traditional Chinese medicine,with a total frequency of 132834 times.The use frequency of rhizoma corydophila,Angelica,Rhizoma corydophila,Motherwort and Ligusticum chuanxiong was in the top,all >2000 times.Most of the drugs used were warm and cold.The medicinal flavor is mainly sweet and spicy;Mostly attributed to liver,spleen two meridians;To adjust chang Qi,invigorate blood as long as the effect.The complex network analysis obtained the core prescriptions: radix cyanoides vinegar,Angelica sinensis,radix cyanoides vinegar,radix paeoniae alba,Ligusticum chuanxiong,salt fennel,Ctyphus officinalis,Motherwort herb,Radix Codonopsis officinalis,fried Fructus briantii with bran,prepared glycyrrhiza glycyrrhiza and the subnetwork diagram of three core drugs.The core compatibility of vinegar myrrh-vinegar rhizoma corydalis,ligusticum chuanxiong-Angelica,vinegar myrrh-Angelica and fried paeoniae paeoniae-angelica was obtained by association rule analysis.The community was divided into 6 categories of prescription: 181 community core Chinese medicine for Chuanxiong,vinegar xiangfu,bran fried fructus aurantii,Sichuan achyranthes bidentata,Motherwort,peony root,cooked rehmannia glutinosa,the main effect is to promote qi and blood circulation and relieve pain;The core traditional Chinese medicine of no.159 club is Black medicine,salt fennel,Bupelum,Angelica,ligusticum chuanxiong,vinegar xiangfu,vinegar myrrh,and its effect is mainly to warm the meridian and remove blood stasis;The core traditional Chinese medicine of the No.4 club is forsythia,coix seed,Cattella,vinegar turtle shell,xylosacea,vinegar fragrant fu,coryma yanhu,with the main effect of clearing heat,promoting blood circulation and relieving pain;No.178 Community core Chinese medicine for vinegar chicken gold,vinegar xiangfu,bran fried fructus aurantii,fried peach kernel,vinegar zedoary,vinegar three ribs,radix scrophulariae,safflower,the effect is mainly to break air and eliminate diseases;The core traditional Chinese medicine of the 166 club is oyster,vinegar fragrant herb,stormweed,Lotus,cooked rehmannia glutinosa,codonopsis pilosula.The core Chinese medicine of no.26 community is cooked rehmannia glutinosa,Coughuang,prepared licorice,Motherwort,Panax notoginseng powder,codonopsis pilosula.The effect is mainly to regulate menstruations and stop bleeding.2.A total of 195 compounds and 307 predicted targets were obtained by retrieving the core prescriptions of Traditional Chinese medicine.223 genes were obtained after being transformed into human genes.Fifty intersected genes were obtained by intersections with 330 genes related to Adenomyosis.The protein-protein interaction network of intersecting genes was constructed and enriched.Finally,four protein functional plates were obtained,including116 items related to biological process,5 items related to cell composition,26 items related to molecular function,and 129 signaling pathways.Conclusion:1.The drugs used to treat adenomyosis are mainly sweet and warm,mostly belonging to liver and spleen meridians;It is mainly effective in activating qi,promoting blood circulation and relieving pain.Complex network analysis obtained the core prescription: vinegar Rhizoma Cyperi,Angelica sinensis,vinegar Rhizoma Corydalis,Radix Paeoniae Alba,Rhizoma Ligustici Chuanxiong,salt fennel,Pollen Typhae,Herba Leonuri,Radix Codonopsis,bran-fried Fructus Aurantii,and Radix Glycyrrhizae Preparata;The community is divided into the first category of core traditional Chinese medicines: Rhizoma Ligustici Chuanxiong,vinegar Rhizoma Cyperi,parched Fructus Aurantii with bran,Radix Cyathulae,Herba Leonuri,Radix Paeoniae Alba,and Radix Rehmanniae Preparata.The main syndrome type of adenomyosis is qi stagnation and blood stasis,and the main treatment is activating blood circulation to remove blood stasis,regulating qi and relieving pain.Warming channels and removing blood stasis,clearing heat and cooling blood and removing blood stasis,breaking qi and eliminating symptoms,nourishing kidney and resolving stagnation,regulating channels and stopping bleeding are the characteristic therapies for adenomyosis.2.Core prescriptions may be involved in hi F-1,PI3K-Akt,FOXO1,estrogen and other signaling pathways to regulate disease progression by affecting VEGFA,MMPs,IL1 B,AKT,ESR and other key targets.It embodies the characteristics of multiple components and multiple targets in TCM treatment of adenomyosis.
