As an important part of Marine traditional Chinese medicine,shell Chinese medicine is not only diverse and abundant in China,but also has a long history of medicinal use and has unique clinical efficacy.Due to the overfishing of local areas and overloaded breeding,as well as technical limitations and ecological environment pollution,the quality of Marine shell Chinese medicinal materials is different.Haliotidis Concha as one of the important Marine shell herbs also have this problem.In addition to the above problems,shell medicinal materials often need to be applied after calcination,but there are key problems such as unclear ination process parameters,imperfect quality standards,identification and market circulation of origin varieties.The existing research literature on the effects of trace elements in Haliotidis Concha is mostly limited to the region,and a few involve origin;there are also few reports on the determination of organic components,such as protein.In order to ensure the integrity and systematization of the study,the Haliotidis Concha purchased in this study is a complete raw shell medicinal material without processing,with the origin and region is defined.Through the quality evaluation of Haliotidis Concha and the composition change of calcination process from the source,aims to find the origin and region’s difference,and further study the calcination process,help to clarify the processing process parameters,to establish more targeted qualitative and quantitative quality control standard to provide research basis,but also hope that the research process of ideas,methods and results for other reference shell Chinese medicine research,lay a foundation for the further comprehensive development and utilization of Marine shell biomedical resources.The study collected Haliotidis Concha’s different region and different origin,By expert identification and using the traditional EDTA-2Na complexation titration method,To determine and compare its calcium carbonate content,So as to achieve the purpose of its preliminary quality evaluation and research;Using a comprehensive spectral analysis technology,namely FTIR technology and XRD technology,Complete the research on the commonness or difference of FTIR characteristic peaks and properties of different origin and different origin;Through trait identification of products and TG-DSC under different calcination conditions,Study the composition change law of the calcination process and clarify the processing process parameters;The first attempt is XRF technology to measure and analyze the content change of trace elements in the stone determination;Using acid formulation to extract protein substances from Haliotidis Concha herbs,And further use of the Brad Ford method to complete the content determination,To compare the content of Haliotidis Concha protein in different region and different origin.On this basis,FTIR technology was extracted through software processing and data integration to measure the characteristic peak and absorbance value,with EDTA-2Na titration method,Ca/Ca O quantitative percentage in XRF technology,and by line chart inflection point comparison method,so as to realize the preliminary exploration of new quantitative method of Haliotidis Concha.The study found that Calcium carbonate(Ca CO3)of Haliotidis Concha’s different region and different origin,The content difference was not obvious.It can be seen that the content parameters of main components alone cannot meet the needs of identification of medicinal materials.But consider studying other components.Different origin and region of Haliotidis Concha medicine,most region and batches of crystal structure stone type and calcite type mixed,mainly stone type.The peaks with higher intensity were concentrated 2θ20°-70°,There are 23angles around 23.11°,26.24°,27.26°,29.44°,31.12°,33.12°,33.14°,36.10°,37.35,37.88°,38.61°,39.44°,41.17°,41.25°,42.93°,45.86°,47.52°,48.54°,50.26°,52.42°,53.02°,57.46°,66.05°.The 2θof the Aragonite are:26.24°,27.26°,31.12°,33.14°,36.10°,37.35,37.88°,38.61°,41.17°,41.25°,42.93°,45.86°,48.54°,50.26°,52.42°,53.02°,66.05°.The2θof the Calcite are 23.11°,29.44°,31.12°,36.10°,39.44°,42.93°,47.52°,48.54°,57.46°,66.05°.Among them,the angles of 33.12°,36.10°,48.54°,and 66.05°are both in the Aragonite and Calcites.However,there are also weak absorption peaks between 2 angles,70°-90°.Considering the number of peaks in this interval,S1-S11 have 29,36,35,37,39,33,38,35,44,32,40 peaks in the range of 2 angles,20°-90°,respectively.The highest peak of Haliotidis Concha medicine is about 29.44°,which represents the Calcium carbonate Calcite,but from the Angle of the crystal.There are 23 XRD diffraction angles of calcium carbonate crystals,including 20 of 2-Theta angles and 10 of 2-Theta angles representing calcite type,four of which have a total of 2-Theta angles.TG-DSC technical analysis showed that the 680±20℃are the temperature ranges of 820±10℃,respectively.The FTIR data of Haliotidis Concha medicine were analyzed macroscopically and did not differ much,and its typical characteristic peak was 3440±20 cm-1、2919±5 cm-1、2520 cm-1、1787 cm-1、1469 cm-1、863±1 cm-1、712 cm-1In consistent with the results in the literature,but more in 699 cm-1Weak absorption peak,which is almost absent in the cannon products.But on the micro level,the origin for Haliotis discus hannai Ino may be better than the origin for Haliotis ruber(Leach),Haliotis laevigata(Donovan)and Haliotis laevigata.Haliotidis Concha in 1787 cm-1The nearby peak is small,and the wave number of the Haliotis ruber(Leach)hull here is 1788cm-1,Consistent with the literature,so the1787 cm-1This is an important breakthrough point in identifying the different origin and authenticity of Haliotidis Concha medicinal herbs;on the other hand,we found 2520cm-1、1787 cm-1/1788 cm-1、1082 cm-1、712 cm-1.It is a relatively stable classic peak that is not easy to shift.The absorbance value of the characteristic peak in FTIR spectroscopy is different between Haliotidis Concha raw and processed products,while the Haliotidis Concha of different origin shows obvious wavenumber displacement in FTIR spectroscopy.This phenomenon provides research ideas for the identification of Haliotidis Concha and other shellfish resources.By comparing the inflection point of the line chart,the method shows that The characteristic calcium carbonate peaks in Ca/Ca O binding to FTIR can be measured using XRF by about 1469cm-1Absorbance values to establish new measurements of calcium carbonate methods.The study found that the absorbance values of the characteristic peak in the FTIR spectrogram were different between Haliotidis Concha unprocessed products and processed products,while the Haliotidis Concha of different origin showed obvious wavenumber displacement in the FTIR spectrogram.As for the research results of other medicinal components of the medicine,the alkaline protein content was about 0.21%,and the trace element content of the Haliotidis Concha was about 2.1%,which contains 19 trace elements.Finally,this study expands the breadth and depth of Haliotidis Concha’s basic research,complements existing research,adds no relevant research data,lays the foundation for quality evaluation and pharmaceutical development,and provides reference for the development of other shellfish resources. |
