Objective:By observing the expression level of TGF-β1,TRAF6 and EMT markers in nasal mucosa of patients with allergic rhinitis(AR),we researched the function and significance of TGF-β 1 and TRAF6 in epithelial mesenchymal transformation of AR.Methods:1.Immunohistochemistry(IHC)was used to examine the expression level of TGF-β 1,TRAF6 and EMT markers(E-cadherin,Vimentin,α-SMA)in nasal mucosa of 15 patients with AR and 15 healthy patients.2.Real-time Quantitative PCR Detecting System(q RT-PCR)was used to examine the m RNA expression level of TGF-β 1,TRAF6 and EMT markers(E-cadherin,Vimentin,α-SMA)in nasal mucosa of 15 patients with AR and 15 healthy patients.3.Western blotting(WB)was used to examine the protein expression level of TGF-β 1,TRAF6 and EMT markers(E-cadherin,Vimentin,α-SMA)in nasal mucosa of 15 patients with AR and 15 healthy patients.4.We analyzed the correlation among TGF-β 1,TRAF6 and EMT markers(E-cadherin,Vimentin,α-SMA)in nasal mucosa of patients with AR.Results:1.The IHC results appeared that the expression level of TGF-β 1,TRAF6,Vimentin and α-SMA in nasal mucosa of AR patients was higher than that of normal controls,and the expression level of E-cadherin in nasal mucosa of AR patients was lower than that of normal controls(P < 0.05).TGF-β 1 protein is mainly expressed on the cell membrane,TRAF6 protein is expressed in the cytoplasm and nucleus,E-cadherin protein is mainly expressed on the cell membrane,and Vimentin protein is mainly expressed in the cell membrane and cytoplasm,α-SMA protein was almost expressed in cytoplasm and nucleus.2.The q RT-PCR results appeared that the expression level of TGF-β1,TRAF6,Vimentin and α-SMA in nasal mucosa of AR patients was higher than that of normal controls,and the expression level of E-cadherin in nasal mucosa of AR patients was lower than that of normal controls(P < 0.05).3.The WB results appeared that the expression level of TGF-β 1,TRAF6,Vimentin and α-SMA in nasal mucosa of AR patients was higher than that of normal controls,and the expression level of E-cadherin in nasal mucosa of AR patients was lower than that of normal controls(P < 0.05).4.TGF-β1 was negatively correlated with the expression of TRAF6(r = 0.8188,P < 0.05).TGF-β1 was negatively correlated with the expression of epithelial cell marker E-cadherin(r =-0.8032,P < 0.05),while TGF-β1 was positively correlated with the expression of interstitial cell markers Vimentin and α-SMA(P < 0.05),and the correlation ratio r were 0.6917 and 0.8076 respectively.There was a negative correlation between TRAF6 and the expression of epithelial cell marker E-cadherin(r=-0.6405,P < 0.05),while TRAF6 was positively correlated with the expression of interstitial cell markers Vimentin and α-SMA(r = 0.5550,P < 0.05;r = 0.5809,P <0.05).Conclusions:1.TGF-β 1,TRAF6 and EMT were highly expressed in nasal mucosa of AR patients.TGF-β 1 and TRAF6 were positively correlated.TGF-β 1 was positively correlated with EMT expression,and TRAF6 was positively correlated with EMT expression.2.TGF-β 1 and TRAF6 may be related to the occurrence of EMT in allergic rhinitis.
