ObjectiveAortic valve calcification disease is increasing year by year,and once it occurs,it will gradually progress,which will seriously affect people’s life and health.At present,it can only be treated by surgery,and the high cost discourages many people.Therefore,it is urgent for researchers to do further research on aortic valve calcification,which is expected to inhibit its progress through drug treatment.This paper will explore the correlation of CCN1 in the process of aortic valve calcification,and find a new breakthrough through bioinformatics research methods.MethodsIn this study,the aortic valves that need to be removed through valve replacement surgery in clinic were collected and divided into 4 cases in the aortic valve calcification group and 4 cases in the non calcification group.The protein CCN1 with biological activity and related to calcification process was finally screened by using isotopic tags for relative and absolute quantification(i TRAQ).t the same time,we conduct bioinformatics analysis,make data statistics from the aspects of subcellular localization,protein domain,go function and KEGG pathway,and obtain significantly changed items through enrichment analysis.Finally,we verified the expression of CCN1 in calcified aortic valves by immunohistochemistry.At the same time,we designed the aortic valve calcification marker OPN as the control experiment to further clarify the correlation between CCN1 and aortic calcification.Results(1)173 key proteins were screened by i TRAQ method,of which 100 differential proteins were up-regulated and 73 differential proteins were down-regulated.Combined with bioinformatics analysis,the protein CCN1 was further studied;(2)Immunohistochemistry Experiments showed that OPN and CCN1 were positively expressed in the calcified group under the microscope,and OPN and CCN1 were negatively expressed under the microscope in the non-calcified group.ConclusionCCN1 may be involved in the regulation of aortic valve calcification.
