| Objective:The expression and related mechanism of WAVE3 and Ki SS-1 in tongue squamous cell carcinoma(TSCC)are not clear.This study detected the expression of WAVE3 and Ki SS-1 in TSCC,combined with clinicopathological data,analyzed the role of WAVE3 and Ki SS-1 in the occurrence,development and prognosis of TSCC,and the correlation between WAVE3 and Ki SS-1 in TSCC,so as to provide ideas and theoretical basis for clinical diagnosis,treatment and prognosis of TSCC patients.Methods:Immunohistochemical staining(IHC)was used to detect the expression of WAVE3 and Ki SS-1 in 38 cases of TSCC and 23 adjacent tissues;Combined with the clinicopathological data and immunohistochemical results of TSCC patients,the factors affecting the prognosis of TSCC patients and their relationship with the overall survival rate of TSCC patients were statistically analyzed;The expression of WAVE3 and Ki SS-1 protein in TSCC was examined by correlation analysis.Results:The positive expression rate of WAVE3 in TSCC was 81.58%(31/38),which was significantly higher than that in adjacent tissues(0/23)(X~2= 38.152 P<0.001)。The positive expression rate of WAVE3 in TSCC was not significantly correlated with age,gender,smoking,drinking,tumor location and T stage(P>0.05),but significantly correlated with lymph node metastasis,tumor clinical stage and tissue differentiation(P < 0.01),and the expression intensity of WAVE3 was also significantly correlated with lymph node metastasis,tumor clinical stage and tissue differentiation(P<0.001).The positive expression rate of Ki SS-1 in TSCC was 23.68%(9/38),while that in adjacent tissues was 8.70%(2/23).There was no significant difference between them(X~2= 1.282 P>0.05)。 The positive expression rate of Ki SS-1 gene was not significantly correlated with age,gender,smoking,drinking,tumor location and T stage(P > 0.05),but significantly correlated with lymph node metastasis,tumor clinical stage and tissue differentiation(P<0.001).The expression intensity of Ki SS-1was significantly correlated with lymph node metastasis,tumor clinical stage and tissue differentiation(P <0.001).Spearman correlation analysis showed that there was a strong negative correlation between WAVE3 and Ki SS-1(r =-0.853,P<0.001).Cox proportional hazards regression model showed that the expression levels of WAVE3 and Ki SS-1,gender,age,smoking,drinking,tumor location,T stage,lymph node metastasis,clinical stage and degree of differentiation had no significant correlation with independent risk prognostic factors of TSCC patients(P>0.05),However,log-rank and Kaplan-Meier analysis showed that the overall survival rate of TSCC patients was related to Ki SS-1 expression,clinical stage and degree of tissue differentiation(P < 0.05),but not to gender,age,smoking,alcohol consumption,tumor location,T stage,lymph node metastasis and WAVE3 expression(P > 0.05).Conclusion:1.WAVE3 and Ki SS-1 are related to the malignant degree of TSCC,suggesting that WAVE3 and Ki SS-1 may be related to the occurrence and development of TSCC.2.Overexpression of WAVE3 and inhibition of Ki SS-1 expression may be important mechanisms for the occurrence,development and metastasis of TSCC.3.The expression levels of WAVE3 and Ki SS-1 can not be used as independent risk prognostic factors related to the overall survival rate of TSCC patients.4.Ki SS-1 may be one of the indicators to evaluate the clinical prognosis of TSCC patients. |
