Objective:Bladder cancer is one of the common malignant tumors of the urinary system.The elderly are prone to bladder cancer because of their low immunity and urinary function degradation.More and more research evidence shows that long non-coding RNA(lnc RNA)plays a key regulatory role in various cancers,which is one of the emerging targets of tumor marker research in recent years.The regulation of lnc RNA on cancer involves many mechanisms and hypotheses,but is closely related to the mechanism of competing endogenous RNA(ce RNA).The regulation of lnc RNA on bladder tumor is a complex process,involving mi RNA,m RNA and other factors.The purpose of this study is to extract the expression data of the three,and use their differential genes to establish a specific ce RNA regulatory network for bladder tumor in elderly patients,and conduct GO and KEGG enrichment analysis.The protein-protein interaction(PPI)network was constructed and survival analysis was performed to find possible biomarkers for elderly bladder tumor and provide basis for diagnosis and treatment.Methods:The data of bladder cancer-related gene expression profile and clinical data of patients aged ≥60 years were downloaded by searching The Cancer Genome Atlas(TCGA)database.Perl software is used to sort out the downloaded gene expression profile data to obtain the gene matrix and match the Ensemble ID.The edg R package of R software was used to analyze the obtained gene matrix,and the differentially expressed lnc RNA,m RNA and mi RNA were screened out.Relevant information was downloaded from mi Rcode,mi RTar Base,Target Scan and mi RDB databases.Perl software and R software were used again to obtain interaction pairs of differentially expressed lnc RNA,m RNA and mi RNA and predict relevant target genes.The obtained interaction pairs were imported into Cytoscape software to draw the regulatory network diagram of ce RNA.Kaplane-meier univariate survival analysis was performed on the differential genes obtained by R software,and the genes related to the prognosis of bladder cancer were obtained.R software was used to analyze the GO and KEGG enrichment maps.Import String online database to build PPI network.Results:A total of 717 lnc RNAs,1819 m RNAs and 158 mi RNAs were significantly differentially expressed in senile bladder tumors.Further,62 lnc RNAs,73 m RNAs and23 mi RNAs were screened for constructing ce RNA network.The obtained differential genes were further processed for survival analysis,and 15 lnc RNAs,7 m RNAs and 3mi RNAs were found to be related to the survival and prognosis of elderly patients with bladder tumor.GO enrichment analysis mainly includes Biological Process(BP)and Cellular Component(CC).KEGG enrichment analysis revealed the related pathways of its concentrated distribution.The PPI network was constructed,and the interaction relationship between proteins corresponding to genes was obtained.Conclusion:In this study,the relevant data in TCGA database were utilized to construct the ce RNA network and obtain 15 Del Ncrnas,7 Demrnas and 3 demirnas closely related to elderly patients with bladder tumor.Most of them have been confirmed to be related to the occurrence,development and prognosis of bladder tumor,but there are still related Rnas in the biological markers of elderly bladder tumor that are worth exploring and studying.Lnc RNA AC025594.2 and PDZRN3-AS1 will be the key research objects in the subsequent stages of this study,to explore the relationship between lnc RNA AC025594.2 and PDZRN3-AS1 with the occurrence,development and prognosis of elderly bladder tumors,so as to clarify their clinical value.
