| Objective:To investigate the effect of silencing LncRNA ANRIL on Ox-LDL-induced apoptosis-associated proteins in human umbilical vein endothelial cells.Methods:Experiment one:1.Give different concentrations(blank control group,25mg/L,50mg/L,100mg/L)Ox-LDL to induce human umbilical vein endothelial cells.2.The expression of ANRIL was detected by RT-qpc R.Experiment two:1.Use lifeter 1.0TM transfection reagent to transfer si ANRIL and si NC into cells,cultivate for 24 hours,and then select the best Ox-LDL(100mg/L)concentration for 24 hours to conduct the experiment.2.RT-qpc R was used to detect the expression of ANRIL in human umbilical vein endothelial cells after transfection of ANRIL interference fragment.3.The relative expression of BAX and BCL-2 was checked by Western blot.Results:1.Compared with the Control group,the expression levels of ANRIL in the Ox-LDL group,Ox-LDL+si NC group and Ox-LDL+si ANRIL group were significantly up-regulated(P<0.05).2.In comparison to Ox-LDL group,the deliver level of ANRIL in control group and Ox-LDL+si ANRIL group decreased significantly(P<0.05),and there was no significant difference in the deliver of ANRIL in Ox-LDL+si NC(P≥0.05).3.Compared with the Ox-LDL+si NC group,the expression level of ANRIL in the Ox-LDL+si ANRIL group was significantly decreased(P<0.05).4.In comparison to Control group,BAX was upregulated and BCL-2 was downregulated in the Ox-LDL group,Ox-LDL+si NC group,or Ox-LDL+si ANRIL group(P < 0.05).5.In the same Ox-LDL group,BAX was down regulated and BCL-2 was up-regulated(P<0.05)in the control,Ox-LDL+si ANRIL groups,while no significant difference was observed in the deliver of BAX and BCL-2 in the Ox-LDL+si NC group(P≥0.05).6.Compared with Ox-LDL+si NC group,the deliver of BCL-2 was up-regulated and BAX was down-regulated in Ox-LDL+si ANRIL group(P<0.05).Conclusion:1.There was a positive correlation between the expression of Ox-LDL and ANRIL,and the two had a correlation.2.Ox-LDL can up regulate the expression amount of ANRIL and make the endothelial cells dysfunctional,leading to the occurrence of as.3.Silencing LncRNA ANRIL made BAX down regulated,BCL-2 up-regulated,alleviated the apoptosis of HUVEC cells induced by Ox-LDL and inhibited the progress of as. |
