Objective: To study the effect of midazolam intraperitoneal injection on bone union of tibial fracture in SD rats.Methods: Forty-eight clean male SD rats(200±20g)were randomly divided into blank group and midazolam group(M group),and the right tibial fracture model was established.On the first day after surgery,rats in M group were intraperitoneally injected with 20mg/kg midazolam,and rats in blank group were intraperitoneally injected with the same dose of normal saline,and 8 rats in each group were sacrificed at the first,second and fourth weeks.During the experiment,the changes of body weight of rats in each group were observed,and the samples to be measured were retained after death.The serum transforming growth factor(TGF)level of rats was determined by ELISA.HE staining was used to observe and evaluate the formation of callus at tibia fracture in rats.The expression of bone morphogenetic protein(BMP)in callus tissue of rat tibial fracture was analyzed by immunohistochemical method.Results: Compared with blank group,serum transforming growth factor(TGF)level of rats in M group was down-regulated(P<0.05);The callus quantity at fracture was decreased in M group(P<0.05);The expression of bone morphogenetic protein(BMP)was down-regulated in M group(P<0.05).Conclusion:Intraperitoneal injection of midazolam can inhibit bone metabolism,down-regulate TGF and BMP levels in rats,and reduce callus quantity at fracture site,suggesting that intraperitoneal injection of midazolam has delayed healing effect on tibial fracture in rats.
