| Objective:By studying the expression of pyroptosis-related genes in gastric cancer and their relationship with prognosis,to construct a pyroptosis-related prognostic model and nomogram to predict the prognosis of gastric cancer patients.And explore the correlation between pyroptosis-related genes and immune cells in gastric cancer.Methods:Transcriptome sequencing data,survival information,and clinical information of gastric cancer patients were downloaded from the TCGA database.Pyroptosis-related genes were obtained from the Human Gene Database.The "LIMMA" R software package and univariate Cox analysis were used to find pyroptosis-related genes with differential expression and prognostic value in the TCGA cohort,and these genes were subjected to GO enrichment and KEGG enrichment analysis.The selected genes were included in multivariate Cox analysis to construct a prognostic model.The patients were divided into high-risk group and low-risk group according to the median risk score,and the predictive value of risk scores for prognosis was evaluated by Kaplan-Meier survival analysis and ROC curve survival analysis.And externally validated on GSE84437 and GSE66229 datasets.Establishment of a nomogram for predicting the survival probability of gastric cancer patients by combining risk scores and patient clinicopathological characteristics.Finally,the correlation between the risk score and immune cell infiltration in patients with gastric cancer was analyzed.Results:1: A prognostic model including 10 genes(BIRC2,CD274,IRGM,ANXA2,GBP5,TXNIP,POP1,GBP1,DHX9,TLR2)was finally established.Compared with the low-risk group,the prognosis of gastric cancer patients in the high-risk group was significantly worse.And it was validated on GSE84437 and GSE66229 datasets.This risk score can be used as an independent prognostic indicator for gastric cancer patients.Low-risk patients had better prognosis in groups >65 years old,≤65 years old,male,female,G1-2,G3,M0,N1-3,I-II,III-IV,and T3-4.2: A nomogram based on risk scores and clinicopathological features can predict the 1-and3-year survival rates of gastric cancer patients.3: Dendritic cells,macrophages,and neutrophils were positively correlated with risk scores.Conclusion:1.We established the prognostic model of pyroptosis related genes for the first time to predict the prognosis of gastric cancer patients.2.The normogram was constructed to predict the prognosis of patients based on pyroptosis related genes,which can accurately predict the 1-year and 3-year survival rates of patients and is of great value for clinical diagnosis and prognosis. |
