Aging has led to a dramatic increase in the incidence of age-related diseases such as Alzheimer’s disease,cardiovascular disease and cancer.There is growing evidence that diet and metabolism play a critical role in determining healthy longevity.The anti-aging effect of natural drugs have garnered increasing interest,and ginseng,as an important part of traditional Chinese medicine,has been widely studied.Although ginseng’s anti-aging properties have been verified in various biological models,the relevant evidence of ginseng on the life span of natural aging model mice is still insufficient.Due to the long lifespan and individual differences of mice,it’s hard to evaluate the anti-aging effect of ginseng by lifespan.Therefore,we chose DNA methylation clock,an objective biomarker,to explore the effect of water extract of ginseng on the methylation age of elderly mice.In addition,we evaluated gene expression levels,intestinal microbiota abundances and behavior measurements of mice,aiming to analyze the regulatory effects of ginseng extract on aging and health in mice through a multi-omics system.DNA methylation sequencing results showed that water extract of ginseng could not significantly change the methylation age of mice,but the DNA methylation age of ginseng group was slightly lower than that of control group at the early stage of feeding.In addition,ginseng could reverse the methylation age of mice to a certain extent,suggesting that ginseng has potential anti-aging effect.The levels of gene expression in mice’s heart tissue were determined,and numerous differentially expressed genes associated with ageing were identified,suggesting that ginseng may improve the immune function of the body by up-regulating or down-regulating the expression of some genes in the heart tissue,such as Cxcl1,Lipe,Isg15 and Cebpa,enhance the immune function of mice,and reduce inflammation symptoms.Analysis of intestinal flora in mice suggests that ginseng may regulate the composition of intestinal species and increase the abundance of probiotics,such as g_Christensenellaceae,g_Bifidobacterium and g_Blauti,which reduce intestinal inflammation and promote intestinal homeostasis.In addition,integrating the transcriptome results,indicates that ginseng may protect cardiomyocytes by interfering with the expression of genes such as Nqo1 and Nampt,which result in the reducing the production of inflammatory factors,protecting myocardial cells,improving function of cardiac muscle contraction in mice,and reducing the risk of age-related cardiovascular diseases such as atherosclerosis.
