| Objective: Sanghuangporus vaninii is a rare edible and medicinal fungus belonging to the genus Sanghuangporus.The fruiting body of Sanghuangporus vaninii is restricted by factors such as low wild yield and long growth cycle,which makes the development and utilization of Phellinus linteus fruiting body.Therefore,it is of great significance to explore the application of Sanghuangporus vaninii liquid fermentation mycelium instead of fruiting bodies.In this experiment,biological fermentation technology was used to optimize the liquid fermentation process of Sanghuangporus vaninii,which provided the technical basis for the large-scale production of Sanghuangporus vaninii mycelium.The chemical composition and antioxidant activity of the mycelium obtained after fermentation were compared with the fruiting bodies of different cultivation substrates,in order to lay a foundation for the application of Sanghuangporus vaninii mycelium instead of fruiting bodies.The therapeutic effect and mechanism of polysaccharides of Sanghuangporus vaninii fermented products against hyperuricemia were studied to provide a new direction for the development and utilization of Sanghuangporus vaninii fermented products.Methods: Single factor investigation,Plackett-Burman experimental design method and Box-Behnken design response surface methodology were used to screen and optimize the Sanghuangporus vaninii liquid fermentation medium and culture conditions.The mycelium obtained by liquid fermentation and the fruiting bodies of different cultivation substrates were tested for the content of moisture,total ash,acid-insoluble ash,extract,crude protein,crude fat and crude fiber according to national standards;by UV-visible spectrophotometry,The contents of total polysaccharides,total flavonoids,total triterpenes,ergosterol and ergosterone were determined by high performance liquid chromatography(HPLC);their antioxidant capacity was evaluated according to DPPH·radical scavenging rate and ABTS+·radical scavenging rate.Male ICR mice were selected to establish a hyperuricemia model,and the therapeutic effect and mechanism of polysaccharides of Sanghuangporus vaninii fermentation products on hyperuricemia mice were investigated.Results: The results of single factor investigation and Plackett-Burman test showed that the most significant factors affecting the yield of intracellular polysaccharide in Sanghuangporus vaninii were carbon source(millet),nitrogen source(soybean meal),and p H;the optimal process was further optimized by response surface methodology.: Carbon source(millet)37 g/L,nitrogen source(bean cake flour)27 g/L,p H 5.7.Moisture,total ash,acid-insoluble ash content: Basswood cultured fruit body >Sawdust cultured fruit body > Mycelium;extract,crude fiber content: Basswood cultured fruit body > Mycelium > Sawdust cultured fruit body;Crude protein,Crude fat content:mycelium > fruit body cultured with basswood > fruit body cultured with wood chips;DPPH·free radical scavenging rate: fruit body cultured with basswood > fruit body cultured with wood chips > mycelium;ABTS+ free radical scavenging rate Rate: Basswood cultured fruiting bodies > mycelium > wood chip cultured fruiting bodies.The results of pharmacological experiments showed that the polysaccharides of Sanghuangporus vaninii fermented products at different doses could reduce the serum UA,CRE,IL-6 and TNF-α content of mice,and the low-dose polysaccharide group of Sanghuangporus vaninii fermented products could reduce the content of XOD;The observation of physical sections showed that the inflammatory cells in liver tissue and the eosinophils in kidney tissue of the polysaccharides of the Sanghuangporus vaninii fermented products were reduced in each dose group;the protein expressions of HMGB1,RAGE and NF-κB p65 in the kidneys of the polysaccharides of the Sanghuangporus vaninii fermented products of each dose group reduce.Conclusion:The optimized fermentation process has stable culture conditions,good repeatability and low cost,which can provide a theoretical basis for the large-scale production of Sanghuangporus vaninii mycelium in the future.Mycelium is expected to be developed and utilized instead of fruiting body;mycelium is higher than fruiting body in the content of total polysaccharide and ergosterol.Application: Pharmacological experiments have shown that the polysaccharides of Sanghuangporus vaninii fermented products have the effect of lowering uric acid,and its mechanism of action is to reduce the production of uric acid by reducing hypoxanthine oxidase(XOD)in the liver,and at the same time downregulate the protein of the cell signaling pathway HMGB1/RAGE/NF-κb expression level,inhibit the release of inflammatory factors,repair liver and kidney damage caused by high uric acid,and restore normal function. |
