Obesity type 2 diabetes mellitus(T2DM)is a common chronic disease,which seriously harms human and animal health.The occurrence of obesity T2 DM is often accompanied by the disorder of calcium and phosphorus metabolism,leading to the occurrence of osteoporosis.Studies have shown that taurine can prevent and cure diabetes and regulate calcium and phosphorus metabolism in osteoporosis laying hens.This test is aimed at by feeding high-fat diet with intraperitoneal injection of chain urea with cephalosporins(STZ)induced obesity T2 DM rats model was established,and add the taurine intervene in the drinking water,discusses taurine prevent obesityⅡ mechanism of action of diabetes complicated with osteoporosis,for the prevention and control of application of taurine calcium phosphorus metabolism disorder caused by obesity T2 DM provide certain theoretical basis.Eight-week-old male Sprague-Dawley rats were randomly divided into three groups: Normal control group(C),model group(M)and taurine intervention group(MT)were pre-fed for 1 week,and the formal experiment was started.Obese T2 DM rat model was prepared by feeding high-fat diet combined with streptozotocin injection,and taurine was added in drinking water for intervention.After 10 weeks of modeling,abdominal aorta blood was collected from the rats,and then the rats were quickly sacrificed.Duodenum,kidney,humerus,femur and tibia tissues of the rats were collected for subsequent tests,and the contents of calcium(Ca),phosphorus(P)and tartrate resistant acid phosphatase(TRAP)in serum were detected.The number of osteoclasts was detected by TRAP staining of paraffin section of the right tibia.Bone mineral density and bone index of humerus,femur and tibia,maximum stress of tibia and bone ash were measured.The m RNA expression levels of calcium and phosphorus metabolization-related factors(VDR,CABP-D28 k,NCX1,PMCA1 b,NAPI-IIb)in duodenum were detected by fluorescence quantitative PCR.Renal calcium and phosphorus reuptake related factors(TRPV5,TRPV6,CABP-D28 k,VDR,PMCA1 b,NCX1,FGFR1,Klotho,Na Pi-IIa,Cyp24a1)m RNA expression levels;MRNA expression levels of bone metabolism related factors(BMP-2,Sphk1,PDGF-bb,ATP6 i,TRAP,Ctsk,MMP-9)in bone tissue.The results showed that after taurine intervention,obese T2 DM rats:(1)Blood Ca and P levels increased significantly;(2)TRAP content decreased significantly,and the number of osteoclasts decreased significantly;(3)Bone mineral density,bone index and tibial maximum stress of humerus,femur and tibia were significantly increased;(4)The expression levels of VDR,CABP-D28 k and NCX1 genes in duodenum were significantly decreased,while the expression levels of PMCA1 b genes were significantly increased.(5)The expression levels of TRPV5,TRPV6,NCX1 and Na Pi-IIa genes were significantly increased,while the expression levels of CABP-D28 k,VDR,PMCA1 b,FGFR1,Klotho and Cyp24a1 genes were significantly decreased.(6)The expression levels of BONE BMP-2,Sphk1 and PDGF-bb were significantly increased,while the expression levels of ATP6 i,TRAP,Ctsk and MMP-9were significantly decreased.The results showed that the supplementation of taurine significantly increased the content of calcium and phosphorus in blood and bone biomechanics in obese T2 DM rats,and the mechanism was related to the promotion of calcium absorption in duodenum,calcium and phosphorus reabsorption in kidney,and the enhancement of bone remodeling.
