Objective: To investigate the expression level and clinical significance of the effect of Lin28 in papillary thyroid carcinoma(PTC)about its impact on the biological behavior of thyroid papillary carcinoma cells and its related regulation mode.Materials and methods: Collected 332 cases of papillary thyroid cancer tissue samples after mastectomy in Shaoxing People’s hospital,and relevant clinical information were compiled.Immunohistochemical method was used to detect the expression of Lin28 in tumor tissues,and the expression and clinical significance of Lin28 protein in thyroid cancer tissues were statistically analyzed.Respectively construct thyroid cancer cell lines overexpressing Lin28 and down-regulating mi R107.Western blotting and RT-QPCR were used to detect the expression of target genes in each group of cells.CCK8 and plate clonogenic assay was used to evaluate cell proliferation.Cell invasion was assessed by scratch experiment,Transwell and Migration experiment.To explore the relationship between Lin28 and Mir-107 and the possible mechanisms.Results: In this study,Lin28 expression level was significantly correlated with lymph node metastasis(P=0.022),but not with gender,age,tumor size,bilateral multiple presence,or extrapapsular thyroid infiltration.Multivariate analysis revealed that Lin28 was associated with lymph node metastasis.Subsequently,we transfected the Lin28 gene in papillary thyroid carcinoma cells and found that the proliferation ability of the transfected Lin28 cells were improved.In the scratch and invasion test,the overexpression of Lin28 could promote the invasion ability of thyroid papillary carcinoma cells,and promote the expression of EMT-related proteins.And Lin28 can inhibit the expression of mi R107.At the same time,decreasing the expression of mi R107 in thyroid papillary carcinoma cells can also promote the proliferation and invasion ability of thyroid papillary carcinoma cells,and significantly increase the expression of Lin28 protein,suggesting that there is an interaction between Lin28 and mi R107.Conclusion: This study found that Lin28 interacts with mi R107 in thyroid papillary carcinoma cells and promotes tumor cell metastasis through the mechanism of epithelial to mesenchymal transition(EMT),thus providing a new theoretical basis and strategy for clinical diagnosis and treatment.
