Objective: Long non-coding RNA(lnc RNA)can affect target genes in a direct or indirect manner,regulating various biological functions including cell proliferation,migration,cycle changes and epigenetics in the human body,which in turn can have a promotive or inhibitory effect on cancer.The aim of this study was to investigate the expression levels of lnc RNA LOC105372270 and gene FBLN5 in pancreatic cancer and the associated clinical significance,with the aim of providing a new direction for the study of molecular mechanisms and therapeutic targets of pancreatic cancer for clinical diagnosis and treatment.Methods:1.The expression of lnc RNA LOC105372270 and gene FBLN5 in pancreatic cancer tissues and corresponding paracancerous pancreatic tissues were detected by q RT-PCR;2.The expression of gene FBLN5 in pancreatic cancer tissues and the corresponding paracancerous pancreatic tissues was detected by protein blotting assay(Western blot);3.The differences in the expression of lnc RNA LOC105372270 and gene FBLN5 in pancreatic cancer tissues were compared within different sub-groups according to the clinical characteristics grouping;4.To detect the expression of lnc RNA LOC105372270 and gene FBLN5 in pancreatic cancer cell lines(PANC-1,HPAF-II)and normal pancreatic ductal epithelial cell lines(HPDE6-C7)by q RT-PCR;5.The expression of lnc RNA LOC105372270 in pancreatic cancer cells PANC-1 was knocked down using si RNA,and CCK-8 was used to detect cell proliferation;6.The expression and characteristics of LOC105372270 in human normal tissues were analyzed by the NCBI database.The subcellular localization of LOC105372270 was predicted by the lnc Locator database;7.The association of LOC105372270 and FBLN5 with clinical characteristics of pancreatic cancer patients was analyzed by Fisher’s exact test.The correlation between the expression levels of LOC105372270 and FBLN5 in pancreatic cancer tissues was analyzed by Spearman’s rank correlation coefficient test.Results:1.The expression levels of LOC105372270 and FBLN5 in pancreatic cancer tissues showed a trend of significant lower expression compared with those in the corresponding paracancerous pancreatic tissues(P < 0.01);2.The expression levels of FBLN5 in translated pancreatic cancer tissues showed a trend of significant low expression compared with those in the corresponding paracancerous pancreatic tissues(P < 0.01);3.The expression levels of LOC105372270 and FBLN5 in pancreatic cancer tissues showed a trend of significantly lower expression in terms of gender(male/female),age(<60 years/≥60 years),CA199(<37U/ml/≥ 37U/ml),CEA(<5ug/L/≥5ug/L),lymph node metastasis(yes/no),TNM stage(I II/III Ⅳ),nerve invasion(yes/no),vascular invasion(yes/no),and site of tumorigenesis(pancreatic head,leptomeninges/body,tail)were not differentially expressed in the subgroups.In the subgroup of differentiation degree(low/medium/high),the expression levels of LOC105372270 in the low differentiation group differed from those in the medium and high differentiation groups,respectively(P < 0.05),and the expression levels of FBLN5 in the low differentiation group differed from those in the medium differentiation group(P < 0.05),which to some extent indicates that the expression of LOC105372270 and FBLN5 in tumor tissues is lower.The lower the expression of FBLN5 in tumor tissues,the more the differentiation of tumor tends to be poorly differentiated this type;4.the expression level of LOC105372270 in pancreatic cancer cell lines(PANC-1,HPAF-II)showed a trend of significantly lower expression compared to that in normal pancreatic ductal epithelial cell lines(HPDE6-C7)(P < 0.05).Similarly,FBLN5 showed a trend of significant low expression in pancreatic cancer cell lines(P < 0.0001);5.The proliferative capacity of the cells was significantly increased after the expression of LOC105372270 was knocked down in pancreatic cancer cells PANC-1(P < 0.001);6.NCBI database sequencing results showed that LOC105372270 was expressed in many normal tissues and organs of human body,and showed differential changes with age.lnc Locator database predicted that LOC105372270 was mainly expressed in the cytoplasm;7.The expression levels of LOC105372270 and FBLN5 in pancreatic cancer tissues had a significant positive correlation(ρ(rho)=0.641;P=0.000),and both were closely correlated with the degree of pathological differentiation of patients,suggesting that our two genes may jointly influence the differentiation of pancreatic tissues and regulate the tumorigenesis and cancer progression of pancreatic cancer through a certain action relationship.Conclusion:The expression levels of Lnc RNA LOC105372270 and FBLN5 were abnormally reduced in pancreatic cancer,and knockdown of LOC105372270 significantly promoted the proliferation ability of pancreatic cancer cells,and the expression levels of this lnc RNA and gene FBLN5 had a significant positive correlation,and their expression levels were closely related to the degree of pathological differentiation,and the lower the expression,the The lower the expression,the worse the pathological differentiation of pancreatic cancer patients.LOC105372270 and FBLN5 have great potential to become effective biomarkers to assist in the diagnosis and treatment of pancreatic cancer,and the specific mechanism of action needs to be further investigated.
