Objectives: To investigate the association between non-alcoholic fatty liver disease and PNPLA3 rs738409 gene polymorphism in Tibetan and Han from Qinghai,China.methods: This study used Case–control study.The study subjects were from the Digestive Departments and Health Examination Center of Qinghai Provincial People’s Hospital.The case group included 152 patients,all patients diagnosed fatty liver by abdominal ultrasound,including 80 Tibetan and 72Han;In addition,135 patients without fatty liver by abnormal ultrasound were selected as the control group,including 61 Tibetan people and 74 Han.The research subjects were surveyed under a questionnaire,conducted a physical examination to collect basic information,drew blood on an empty stomach to detect biochemical indicators and for PCR amplification to detect genotype.Logistics regression analysis was used to analyze the association between PNPLA3 rs738409 gene polymorphism and non-alcoholic fatty liver disease in Tibetan and Han nationality in Qinghai.Results:In the study population,the distribution of genotype frequency of CC,CG,GG is 24(18.46%),48(36.92%),58(44.62%)respectively in the case group,and the distribution of genotype frequency of CC,CG,GG in the control group is 43(64.2%),42(46.7%),21(26.6%)respectively,GG,GG genotype frequency in the case group was obviously higher than that of control group(p=0.000).The distribution of allele frequency of C allele and G allele are 6(36.93%),164(63.07%)respectively in the case group,the C allele and G allele are 128(60.38%),84(39.62%)respectively in the control group,and G allele frequency in the case group was obviously higher than that of control group(p=0.000).The NAFLD risk factors’binary logistics analysis shows,carrying CC genotype,the CG and GG genotypes can increase the risk of NAFLD in the Han nationality in Qinghai,the Odds ratio(OR)and a 95% confidence interval(CI)is 3.210(1.388~7.420);Carrying CC genotype,The CG and GG genotypes can increase the risk of developing NAFLD in the Tibetan nationality in Qinghai,the OR is 3.068,95%CI(1.217~7.735);Carrying C allele,the G allele can increase the risk of developing NAFLD in the Han nationality in Qinghai,The OR is 3.215,95%CI(1.798~5.748).Carrying the C allele,the G allele can increase the risk of developing NAFLD in the Tibetan nationality in Qinghai,the OR is 2.656,95%CI(1.452~4.860).Conclusions: Among the Han and Tibetan nationality in the Qinghai region,The PNPLA3 rs738409 gene polymorphism in the population with NAFLD was dominated by CG and GG types;The PNPLA3 rs738409 genotype can increase the risk of NAFLD with the GG genotype among the Han and Tibetan nationality in the Qinghai region;Carrying the G allele of the PNPLA3rs738409 gene increases the risk of NAFLD among the Han and Tibetan nationality in the Qinghai region. |
