Objective:To analyze the effect of sodium hydrosulfide on the expression of 6-keto-PGF1α、v WF 、s LOX-1 of vascular endothelial cells after the intervention of rat burn serum and the rate of cell inhibition,and then to explore the effect of sodium hydrosulfide on vascular endothelial cells after burn,and to study the treatment of body after burn from the cellular level.Methods:SD rats were treated with simulated burns and burn modeling.Blood was taken from abdominal aorta and centrifuged to obtain serum for standby.After the passage to the third generation of rat vascular endothelial cells was synchronized,10%simulated burn serum and 10% burn serum culture medium were intervened,and then150 μmol/L sodium hydrosulfide(the optimal concentration of cell inhibition test results)solution was divided into groups for intervention experiment,and the cell supernatant was collected.The experiment was divided into three groups:simulated burn rat serum intervention group;burn rats serum intervention group;burn rat serum +sodium hydrosulfide intervention group.The cell inhibition rate was detected by cell count detection kit-8 method(CCK-8),enzyme linked immunosorbent assay(ELisa)was used to detect 6-keto-PGFlα、v WF and s LOX-1 in endothelial cell supernatant.Results:The cell inhibition rate of rats under the intervention of 150μmol/L sodium hydrosulfide was the lowest.(1)6-keto-PGFlα in endothelial cell supernatant of burn serum group,the concentration of was significantly higher than that in the supernatant of endothelial cells in the simulated burn serum group.the difference was statistically significant(P<0.05);6-keto-PGFlα in endothelial cell supernatant of burn serum + Na HS group,the concentration of 6-keto-PGFlα was lower than that in endothelial cell supernatant of burn group,the difference was statistically significant(P<0.05).(2)The concentration of v WF in endothelial cell supernatant of burn serum group was higher than that of simulated burn serum group,the difference was statistically significant(P<0.05);The concentration of v WF in endothelial cell supernatant in burn serum + Na HS group was lower than that in burn serum group,the difference was statistically significant(P<0.05).(3)The concentration of s LOX-1 in endothelial cell supernatant of burn serum group was higher than that of simulated burn serum group,the difference was statistically significant(P<0.05);The concentration of s LOX-1 in endothelial cell supernatant in burn serum + Na HS group was lower than that in burn serum group,the difference was statistically significant(P<0.05).Conclusion:(1)Exogenous sodium hydrosulfide can induce 6-keto-PGFlα in vascular endothelial cells of burned rats decreased expression;(2)Exogenous sodium hydrosulfide can reduce the expression of v WF in vascular endothelial cells of burned rats;(3)Exogenous sodium hydrosulfide can reduce the expression of s LOX-1 in vascular endothelial cells of burned rats;(4)The coagulation function of vascular endothelial cells in burned rats can be affected by exogenous sodium hydrosulfide.
