Effect Of Wenyang Yiqi Huoxue Prescription On Myocardial Myosin And Troponin In Rats With Chronic Heart Failure

Objective:The effect of Wenyang Yiqi Invigorating Blood Formula on myocardial myoglobulin heavy chain,regulatory light chain and troponin in rat models of chronic heart failure was observed,and the possible mechanism of action of this formula in the treatment of chronic heart failure was discussed,which provided experimental basis for further research and use of this formula.Methods:Modelding was made by subcutaneous injection of isoproterenol hydrochloride,and successful moulding was randomly divided into model group,western medicine group,wenyang qi and low,medium and high dose group.Rats received different drugs(10ml·Kg^-1).The BNP levels were determined by cardiac ultrasound and RT-PCR for myosin Myh6,Myh7 and troponin cTnT mRNA,MHC and MLC2 and troponin cTnT protein in cardiac tissue of chronic heart failure.Results:1.Compared with the blank group,the heart ejection fraction of rats decreased（P<0.01）;the serum BNP level increased（P<0.01）;proved the success of chronic heart failure model.2.Compared with the blank group,in the model group,the expression of Myh6mRNA,regulatory light chain MLC2,and cTnT in cardiac muscle decreased（P<0.05）and Myh7 mRNA expression increased in the heavy chain（P<0.05）.Compared with the model group,the Myh7 mRNA expression in the myocardial tissue in the group（P<0.05）,and the MLC2 and troponin cTnT increased（P<0.01）between the Myh6mRNA and troponin cTnT mRNA（P>0.05）.Conclusion:Studies have confirmed that the possible mechanism of action of Wenyang Yiqi Invigorating Blood Formula for the treatment of chronic heart failure is to inhibit the loss of troponin cTnT in myocardial tissue and the expression of Myh7mRNA protein in the heavy chain of myoglobulin,and improve the expression level of myocardial myoglobulin regulatory light chain MLC2 protein;and its efficacy has a positive correlation between the drug concentration.
