| Objective:Because of the occult incidence of lihepatocellular carcinoma,most patients are in the middle and late stage at the time of diagnosis,and their five-year survival rate is less than 20%.RBPs are involved in the occurrence of hepatocellular carcinoma through a variety of mechanisms,and play an important role in the prognosis of patients with hepatocellular carcinoma.Therefore,this study used RBPs to construct a hepatocellular carcinoma prognosis evaluation model to evaluate the prognosis of patients with hepatocellular carcinoma.Methods:Download the expression profile data and clinical data of hepatocellular carcinoma samples and normal liver tissue samples from TCGA database and ICGC database.Differentially expressed RBPs were screened by differential analysis.Go and KEGG enrichment analysis were carried out to explore the function of differential RBPs.PPI network was constructed by differentially expressed RBPs and hub genes were screened.Cox regression analysis was used to screen RBPs related to the prognosis of hepatocellular carcinoma,and they were used to construct the prognosis model of hepatocellular carcinoma.Finally,the effectiveness of the model is evaluated.Results:There were 135 differentially expressed RBPs,including 113 high expression and 22 low expression.Enrichment analysis showed that differential RBPs were mainly enriched in RNA metabolic regulation(RNA splicing,RNA degradation and RNA transport),DNA methylation and so on.The six hub genes in PPI network play an important role in the pathogenesis of hepatocellular carcinoma.DCAF13,PURG,ADAD1 and EZH2 are related to the prognosis of hepatocellular carcinoma and are used to construct the prognosis model of hepatocellular carcinoma.The risk score can be used as an independent prognostic factor for the prognosis of hepatocellular carcinoma.Conclusions:DCAF13,PURG,ADAD1 and EZH2 are significantly overexpressed in hepatocellular carcinoma,which is closely related to the poor prognosis of hepatocellular carcinoma.The prognosis model of hepatocellular carcinoma based on DCAF13,PURG,ADAD1 and EZH2 is helpful to the prognosis evaluation of hepatocellular carcinoma and guide clinical decision-making and treatment.T stage and risk score are significantly correlated with the prognosis of patients with hepatocellular carcinoma,and the risk score can be used as an independent prognostic factor of hepatocellular carcinoma. |
