| [Purpose]This study summarizes the diagnosis and treatment of hyperuricemia in traditional Chinese medicine and western medicine through a literature review and then analyzes the rules of traditional Chinese medicine for hyperuricemia based on data mining.Finally,network pharmacology was used to explore the role of Luo’s gout prescription in the diagnosis and treatment of hyperuricemia and verified by molecular docking technology.[Materials and methods]1.Data mining:By searching the four major databases of VIP,CNKI,Biomedicine,and Wanfang,the clinical literature on the treatment of hyperuricemia with traditional Chinese medicine was published from the establishment to February 1,2022,and screened according to the inclusion and exclusion criteria.Then the included literature conducted descriptive analysis and the prescriptions were extracted,normalized,and split,which was converted into data forms suitable for statistical analysis.Finally,the frequency analysis,association analysis,clustering rule analysis,and complex network analysis of high-frequency drugs were carried out with the help of the ancient and modern medical record cloud platform software.2.Network pharmacology and molecular docking:The TCMSP and ETCM databases were used to screen the active ingredients and targets of Luo’s gout prescription.The Genecards database was used to screen the disease targets of hyperuricemia.Then the DAVID database performed GO and KEGG analysis on the common targets.The protein interaction(PPI)analysis of the intersection targets was carried out using the String platform,and a visual network diagram was constructed with the help of Cytoscape 3.6.0 software to screen out the core active components and key targets.Finally,the molecular docking verification was carried out using software such as Auto Dock Vina and PyMOL.[Results]1.The screening results were 216 clinical documents,208 effective prescriptions,242 traditional Chinese medicines,and the frequency of occurrence of the drugs involved 2393 times.The top three traditional Chinese medicines most frequently used are Tuckahoe,Coix Seed,and Achyranthes.12 commonly used combinations are obtained through association rule analysis,and the normal one is Bixie-Tuckahoe.Four groups of commonly used classifications were obtained by cluster analysis,among which Atractylodes Rhizoma and Cortex Phellodendri had the highest homogeneity.Six core drug combinations were obtained by complex network analysis.2.294 active ingredients,711 drug targets,and 692 disease genes related to hyperuricemia were obtained from Luo’s gout prescription.There were 131 targets in common between prescriptions and diseases,of which 626 targets in GO analysis(P<0.05),involving estradiol,lipopolysaccharide,etc.,and 137 targets in KEGG analysis(P<0.05),involving IL-17 and HIF-1 signaling pathway,etc.The top 2 core active ingredients are quercetin and stigmasterol.The top 2 key targets are GAPDH and INS.Taking Benzbromarone as the control group,the binding efficacy between traditional Chinese medicine and western medicine to the core target was compared at the molecular docking level.The results showed that the active ingredients of Luo’s gout prescription had a better combination with the core targets.[Conclusions]1.In the treatment of hyperuricemia with traditional Chinese medicine,the drugs are mostly flat,cold,and warm.The five flavors are sweet,bitter,and light and the meridians belong to the liver,spleen,and stomach meridians.Prescription drugs mostly choose heat-clearing drugs,diuresis-dampening drugs,and tendons-strengthening drugs.Tuckahoe-Bixie is the core drug pair.2.The network pharmacology method was used to preliminarily predict the target and action pathway of the Luo’s gout prescription in the treatment of hyperuricemia,which provides a theoretical basis for further experimental research of this formula. |
